異甘草素誘導(dǎo)人宮頸癌細(xì)胞增殖的影響及作用機(jī)制的研究

【摘要】:目的研究異甘草素(ISL)抑制宮頸癌Hela細(xì)胞增殖作用及促進(jìn)細(xì)胞凋亡的作用機(jī)制。方法采用0.025、0.050、0.100、0.200、0.300、0.400mg/mL濃度ISL處理HeLa細(xì)胞,采用四甲基偶氮唑鹽(MTT)比色法檢測HeLa細(xì)胞增殖;采用流式細(xì)胞儀檢測HeLa細(xì)胞凋亡情況;應(yīng)用羅丹明123染色法檢測HeLa細(xì)胞線粒體跨膜電位的變化;采用RT-PCR檢測6種不同濃度ISL對Bcl-2、P53、P21、E6mRNA基因表達(dá)的影響。結(jié)果 (1)MTT檢測結(jié)果顯示:隨著ISL濃度增加,對不同時間段HeLa細(xì)胞的抑制作用明顯增加(P0.05);(2)流式細(xì)胞儀檢測:相比于空白對照組,隨著ISL濃度逐漸增加,Hela細(xì)胞凋亡率明顯升高(P0.01),且凋亡率呈劑量依賴性;(3)羅丹明123熒光染色檢測結(jié)果:隨著ISL濃度逐漸增加,細(xì)胞線粒體膜電位下降(P0.01);(4)RT-PCR結(jié)果顯示:隨著ISL濃度逐漸增加,E6、Bcl-2mRNA表達(dá)水平明顯降低(P0.05),而P53、p21mRNA表達(dá)水平越升高(P0.05)。結(jié)論 ISL通過下調(diào)E6、Bcl-2的表達(dá)和上調(diào)P21、P53基因的表達(dá)來抑制宮頸癌細(xì)胞的增殖。
[Abstract]:Objective to study the mechanism of isoliberrrhizin (ISL) inhibiting the proliferation and promoting apoptosis of cervical cancer Hela cells. Methods HeLa cells were treated with 0.025 ~ 0.050 ~ 0.100 ~ 0.200 ~ 0.200 mg / mL ISL, the proliferation of HeLa cells was detected by MTT colorimetry, the apoptosis of HeLa cells was detected by flow cytometry, the changes of mitochondrial transmembrane potential of HeLa cells were detected by Rhodamine 123 staining method. The effect of six different concentrations of ISL on the expression of Bcl-2P53P53P51P21 / E6 mRNA was detected by RT-PCR. Results: with the increase of ISL concentration, the inhibitory effect on HeLa cells at different time periods was significantly increased by flow cytometry. With the increase of ISL concentration, the apoptotic rate of Hela cells increased significantly, and the apoptosis rate was dose-dependent. The results of Rhodamine 123 fluorescence staining showed that: with the increase of ISL concentration, the apoptosis rate of Hela cells increased with the increase of ISL concentration in a dose-dependent manner. The results of RT-PCR showed that with the increase of ISL concentration, the expression level of Bcl-2 mRNA decreased significantly, while the expression level of P53 P21 mRNA increased significantly. Conclusion ISL inhibits the proliferation of cervical cancer cells by down-regulating the expression of E6, Bcl-2 and up-regulating the expression of P21 ~ + -p53 gene.
