原因不明復(fù)發(fā)性流產(chǎn)患者外周血單核細胞上Tim-3和PD-1的表達及意義

【摘要】:目的探討原因不明復(fù)發(fā)性流產(chǎn)(unexplained recurrent spontaneous abortion,URSA)和先兆流產(chǎn)患者外周血單核細胞上PD-1和Tim-3的表達及其細胞因子的分泌情況,為先兆流產(chǎn)和URSA發(fā)病機制研究提供理論依據(jù)。方法隨機選取29例健康早孕婦女作為正常早孕組(A組),37例僅有少量陰道出血的早孕婦女為先兆流產(chǎn)組(B組),22例URSA早孕患者為URSA組(C組);應(yīng)用密度梯度離心法分離外周血單個核細胞,然后采用流式細胞分析技術(shù)檢測3組孕婦外周血中CD14+單核細胞上Tim-3、PD-1的表達水平;ELISA法檢測外周血中IL-4、IL-12、IFN-γ的含量。結(jié)果與正常早孕組相比,先兆流產(chǎn)組和URSA組外周血單核細胞上Tim-3、PD-1表達降低,外周血IL-4表達降低,URSA組低于先兆流產(chǎn)組(P0.05);IFN-γ、IL-12表達增高,URSA組高于先兆流產(chǎn)組(P0.05);相關(guān)性分析提示先兆流產(chǎn)組及URSA組外周血單核細胞上Tim-3與PD-1之間呈正相關(guān);Tim-3、PD-1與IL-12呈負相關(guān),IL-12與IL-4呈負相關(guān),與IFN-γ呈正相關(guān)。結(jié)論外周血單核細胞上Tim-3、PD-1可能通過調(diào)節(jié)IL-12產(chǎn)生間接調(diào)節(jié)Th1/Th2平衡參與先兆流產(chǎn)和URSA的發(fā)生。
[Abstract]:Objective to investigate the expression of PD-1 and Tim-3 and cytokine secretion in peripheral blood monocytes of patients with unexplained recurrent spontaneous abortion (Ursa) and threatened abortion, and to provide theoretical basis for the study of pathogenesis of threatened abortion and URSA. Methods Twenty-nine healthy early pregnant women were randomly selected as normal early pregnancy group (group A), 37 early pregnant women with only a small amount of vaginal bleeding were selected as threatened abortion group (group B) and 22 patients with URSA early pregnancy as URSA group (group C), and were separated by density gradient centrifugation. Peripheral blood mononuclear cells, Then the expression level of Tim-3P PD-1 on CD14 monocytes in three groups of pregnant women was detected by flow cytometry and the level of IL-4 IL-12 IFN- 緯 in peripheral blood was detected by Elisa. Results compared with normal early pregnancy group, the expression of Tim-3 PD-1 on peripheral blood monocytes in threatened abortion group and URSA group was decreased. The decrease of IL-4 expression in peripheral blood in Ursa group was lower than that in threatened abortion group and the expression of IFN- 緯 -IL-12 in Ursa group was higher than that in threatened abortion group, and the correlation analysis indicated that there was a positive correlation between Tim-3 and PD-1 in peripheral blood monocyte of threatened abortion group and URSA group. There was a negative correlation between IL-12 and IL-4. There was a positive correlation with IFN- 緯. Conclusion Tim-3 PD-1 may be involved in the occurrence of threatened abortion and URSA by regulating the production of IL-12 and indirectly regulating the balance of Th1/Th2 in peripheral blood monocytes.
