【摘要】：目的探討腺苷A1受體(A1R)在妊娠期高血壓以及子癇前期發(fā)病機(jī)制中的作用。方法取剖宮產(chǎn)分娩的妊娠期高血壓(n=6)、子癇前期(n=14)和正常對(duì)照(n=15)產(chǎn)婦胎盤(pán)組織,采用免疫熒光檢測(cè)A1R在胎盤(pán)組織中的表達(dá)部位,蛋白質(zhì)印跡法檢測(cè)A1R在胎盤(pán)中的表達(dá)量。建立常氧、缺氧(8、24、48、72h)以及缺氧復(fù)氧(8h)滋養(yǎng)細(xì)胞模型,運(yùn)用實(shí)時(shí)熒光定量PCR和蛋白質(zhì)印跡法檢測(cè)A1R在細(xì)胞中的表達(dá)情況,Transwell和蛋白質(zhì)印跡法檢測(cè)加入A1R拮抗劑8-環(huán)戊基-1,3-二丙基黃嘌呤(DPCPX)后對(duì)缺氧的胎盤(pán)滋養(yǎng)細(xì)胞侵襲性和蛋白激酶A(PKA)表達(dá)的影響。結(jié)果 A1R在胎盤(pán)表面的滋養(yǎng)細(xì)胞以及內(nèi)皮細(xì)胞中均有表達(dá)。與正常胎盤(pán)比較,A1R在輕度和重度子癇前期胎盤(pán)組織中表達(dá)升高(P0.01)。A1R在體外胎盤(pán)滋養(yǎng)細(xì)胞缺氧模型中的表達(dá)隨著缺氧時(shí)間的延長(zhǎng)而逐漸升高(P0.05)。胎盤(pán)滋養(yǎng)細(xì)胞缺氧模型中滋養(yǎng)細(xì)胞的侵襲能力較正常組下降(P0.01),PKA表達(dá)下降(P0.05);加入拮抗劑DPCPX可以增強(qiáng)滋養(yǎng)細(xì)胞的侵襲性(P0.05),同時(shí)上調(diào)PKA的表達(dá)水平(P0.05)。結(jié)論缺氧程度的增加可以使A1R表達(dá)升高,A1R在胎盤(pán)滋養(yǎng)細(xì)胞中的作用機(jī)制可能與調(diào)節(jié)PKA相關(guān)。
[Abstract]:Objective to investigate the role of adenosine A 1 receptor A 1 R in the pathogenesis of hypertension complicating pregnancy and preeclampsia. Methods the placenta tissues of pregnant women with hypertension complicating pregnancy during cesarean section were collected. The expression of A1R in placenta was detected by immunofluorescence, and the expression of A1R in placenta was detected by Western blot. The model of trophoblastic cells was established with normoxic oxygen, anoxic anoxia and anoxic reoxygenation for 72h) and anoxia reoxygenation for 8h). The expression of A1R in human placental trophoblast cells was detected by real-time fluorescence quantitative PCR and Western blotting. Transwell and Western blotting were used to detect the invasiveness of placental trophoblastic cells induced by hypoxia by adding 8-cyclopentyl-1-dipropylxanthine 3- dipropylxanthine into A1R antagonist. And protein kinase (PKA) expression. Results A 1R was expressed in trophoblast and endothelial cells. Compared with normal placenta, the expression of A1R in placenta of mild and severe preeclampsia was higher than that of normal placenta. The invasion ability of trophoblast in placental trophoblastic hypoxia model was lower than that in normal group, and the expression of P0.05A was decreased in placental trophoblastic hypoxia model, and the level of PKA expression was up-regulated by adding antagonist DPCPX to enhance the invasiveness of trophoblast. Conclusion the increase of hypoxia may increase the expression of A1R in placental trophoblastic cells and its mechanism may be related to the regulation of PKA.
