卵巢衰老中細(xì)胞衰老現(xiàn)象初探

【摘要】:目的:第一部分:為了探究細(xì)胞衰老與卵巢衰老的關(guān)系,采用衰老相關(guān)的半乳糖苷酶(Senescence Associated b-galactosidase, SA-b-gal)染色方法觀察在不同年齡階段的成年C57BL/6J雌鼠卵巢組織細(xì)胞衰老現(xiàn)象的情況,探索在卵巢生理性衰老過程中,是否存在細(xì)胞衰老(cellular senescence)的現(xiàn)象,探索其在卵巢組織各類細(xì)胞中的分布,同時(shí)觀察細(xì)胞衰老的程度隨著年齡增長的變化; 第二部分:為了探究細(xì)胞衰老與卵泡早期發(fā)育的關(guān)系,本課題檢測了新生及幼齡C57BL/6J雌鼠卵巢組織中細(xì)胞衰老的情況,探索在卵泡發(fā)育過程中細(xì)胞衰老現(xiàn)象的存在、分布以及變化。 通過以上兩個(gè)方面闡明了細(xì)胞衰老與卵巢器官衰老、卵泡發(fā)育閉鎖之間的關(guān)系。 方法:第一部分選取6周、12周、32周、40周、48周年齡階段C57BL/6J雌性小鼠,檢測各年齡段小鼠卵巢組織中衰老相關(guān)的半乳糖苷酶活性情況;第二部分選取2天、5天、7天及3周年齡階段C57BL/6J雌性小鼠,檢測各年齡段小鼠卵巢組織中衰老相關(guān)的半乳糖苷酶活性情況。 結(jié)果:通過衰老相關(guān)半乳糖苷酶活性檢測法發(fā)現(xiàn)卵巢組織間質(zhì)中,細(xì)胞衰老現(xiàn)象隨著年齡的增長而增加,而在卵泡膜細(xì)胞中未見明顯表達(dá),顆粒細(xì)胞中未發(fā)現(xiàn)明顯的增齡性趨勢(shì);在雌性小鼠出生后的卵泡早期發(fā)育過程中,未見明顯細(xì)胞衰老現(xiàn)象出現(xiàn)在任何細(xì)胞中。 結(jié)論:在卵巢衰老過程中,間質(zhì)細(xì)胞的衰老占據(jù)主要部分,并呈現(xiàn)隨著年齡逐漸增長的趨勢(shì),在成年卵巢組織顆粒細(xì)胞細(xì)胞衰老現(xiàn)象未見明顯增齡性趨勢(shì),而卵泡膜細(xì)胞未見衰老表現(xiàn);在新生期卵泡早期發(fā)育過程中,未見細(xì)胞衰老現(xiàn)象。
[Abstract]:Objective: to investigate the relationship between cell senescence and ovarian senescence, senescence Associated b-galactosidase (SA-b-galgalase) staining was used to observe the aging of ovarian tissue cells in adult C57BL/6J female rats at different ages. To explore whether there is cellular senescence in the process of ovarian physiological senescence, to explore the distribution of cellular senescence in all kinds of ovarian cells, and to observe the change of cell senescence with age. The second part: in order to explore the relationship between cell senescence and early follicle development, we detected the cell senescence in ovarian tissue of newborn and young C57BL/6J mice, and explored the existence of cell senescence during follicular development. Distribution and variation. The relationship between cell senescence and ovarian organ senescence and follicular development atresia was clarified. Methods: in the first part, the aging related galactosidase activities in the ovarian tissues of C57BL/6J female mice were measured at the age of 6 weeks, 12 weeks, 32 weeks, 40 weeks and 48 weeks. In the second part, the senescence related galactosidase activity in ovarian tissues of C57BL/6J female mice of 2 days, 5 days, 7 days and 3 weeks was determined. Results: through the assay of senescence related galactosidase activity, it was found that cell senescence increased with age, but not in follicular membrane cells. There was no obvious aging trend in granulosa cells and no obvious cell senescence was found in any of the early follicles of female mice during the early development of postnatal follicles. Conclusion: in the process of ovarian senescence, the senescence of stromal cells occupies the main part and tends to increase with age, but there is no obvious aging trend in granulosa cells of adult ovarian tissue. However, there was no senescence in the follicular membrane cells and no senescence in the early development of the newborn follicles.
