【摘要】：目的： 檢測細(xì)胞周期素E（CyclinE）、PTEN在上皮性卵巢癌的表達，分析其與上皮性卵巢癌臨床病理因素關(guān)系及二者表達的相關(guān)性，探究其在腫瘤發(fā)生發(fā)展中的功用與關(guān)聯(lián)，明了其在上皮性卵巢癌診斷、預(yù)后等方面的應(yīng)用。 方法： 選用鏈霉素—過氧化物酶(S-P)免疫組化方法。檢測CyclinE和PTEN在正常卵巢組織、良性上皮性卵巢腫瘤組織、交界性上皮性卵巢腫瘤組織和上皮性卵巢癌組織中的表達情況。應(yīng)用SPSS19.0軟件包進行統(tǒng)計學(xué)分析。采用χ2檢驗和Fisher確切概率法分析CyclinE和PTEN在各組織中的表達及與臨床病理因素之間的關(guān)系；采用Spearman相關(guān)分析二者的相互關(guān)聯(lián)性。 結(jié)果： 1.CyclinE在正常卵巢、卵巢良性腫瘤、卵巢交界性腫瘤、卵巢上皮性癌的表達率分別為5%，14.3%，40.6%，67.5%，呈不斷上升趨勢，四組之間差異顯著(p0.001)。正常組、良性組CyclinE蛋白的表達明顯低于惡性組，差別顯著(P0.001)；交界性組、惡性組之間比較，差異有統(tǒng)計學(xué)意義(P0.05)；正常組與交界性組、良性組與交界性組之間比較，差別顯著(P0.05)；但正常組與良性組，差異無統(tǒng)計學(xué)意義(P0.05)。 2.在上皮性卵巢癌患者中，高中分化患者腫瘤組織中CyclinE蛋白表達陽性率低于低分化者，，有統(tǒng)計學(xué)差異(p0.05)；FIGO分期Ⅲ～Ⅳ患者腫瘤組織中CyclinE蛋白表達陽性率高于Ⅰ～Ⅱ期患者，有統(tǒng)計學(xué)差異(p0.05)；有淋巴結(jié)轉(zhuǎn)移的患者腫瘤組織中CyclinE蛋白表達陽性率高于無淋巴結(jié)轉(zhuǎn)移患者，差異顯著(p0.05)。上皮性卵巢癌CyclinE的表達與年齡、組織學(xué)類型均無關(guān)。 3.PTEN在正常卵巢、良性卵巢腫瘤、交界性卵巢腫瘤和上皮性卵巢癌中表達率依次為100%，85.70%，81.30%，40%，呈逐步降低趨勢，四組之間差異顯著(p0.001)。正常組、良性組二者PTEN的表達明顯高于惡性組，差別顯著(P0.001)；交界性組、惡性組之間比較，差異有統(tǒng)計學(xué)意義(P0.001)；但正常組與良性組、正常組與交界性組、良性組與交界性組之間，無顯著差異(P0.05)。 4.在上皮性卵巢癌患者中，高、中分化患者的腫瘤組織中PTEN表達陽性率高于低分化者，有統(tǒng)計學(xué)差別(p0.05)；FIGO分期Ⅲ～Ⅳ患者腫瘤組織中PTEN表達陽性率比Ⅰ～Ⅱ期患者低，有統(tǒng)計學(xué)差別(p0.05)；淋巴結(jié)有轉(zhuǎn)移組低于無轉(zhuǎn)移組，有顯著差異(P0.05)。PTEN的表達與患者的年齡、組織學(xué)類型無相關(guān)性。 5.在上皮性卵巢癌中，CyclinE、PTEN的表達呈負(fù)相關(guān)(rs=-0.414，P0.01)。 結(jié)論： 1.CyclinE在正常卵巢組織、良性卵巢腫瘤、交界性卵巢腫瘤及上皮性卵巢癌中的表達，呈逐漸上升的趨勢。隨著FIGO分期的增高，卵巢上皮性癌組織中CyclinE表達上升；隨著病理分級的下降，上皮性卵巢癌組織中CyclinE表達上升；有淋巴結(jié)轉(zhuǎn)移的患者CyclinE的表達明顯升高。結(jié)果表明，CyclinE在卵巢上皮性癌的發(fā)生和發(fā)展中起重要促進作用。 2.PTEN在正常卵巢組織、良性卵巢腫瘤、交界性卵巢腫瘤及上皮性卵巢癌中的表達，呈逐漸下降的趨勢。隨著FIGO分期的增加，卵巢上皮性癌組織中PTEN表達降低；隨著病理分級的下降，上皮性卵巢癌組織中PTEN表達減少；有淋巴結(jié)轉(zhuǎn)移的患者PTEN的表達明顯下降。結(jié)果表明，PTEN在卵巢上皮性癌的發(fā)生和發(fā)展中起重要抑制作用。 3.CyclinE和PTEN在上皮性卵巢癌中的表達呈負(fù)相關(guān)。兩者之間可能存在協(xié)同作用，聯(lián)合檢測對上皮性卵巢癌的診斷、預(yù)后有重要的參考價值。
[Abstract]:Purpose :

The expression of cyclin E ( Cyclin E ) and PTEN in epithelial ovarian carcinoma was examined . The correlation between the expression of cyclin E and PTEN in epithelial ovarian carcinoma was analyzed . The utility and association of the expression of cyclin E and PTEN in the development of epithelial ovarian cancer were investigated .

Method :

The expression of Cyclin E and PTEN in normal ovarian tissues , benign epithelial ovarian tumor tissues , borderline epithelial ovarian tumor tissues and epithelial ovarian cancer tissues was investigated by SPSS 10.0 software . The expression of Cyclin E and PTEN in various tissues and their relationship with clinicopathological factors were analyzed by 蠂2 test and Fisher exact probability method .
The correlation between the two correlations was analyzed by using the correlation analysis between the two methods .

Results :

1 . The expression rates of Cyclin E in normal ovary , benign ovarian tumor , borderline ovarian tumor and epithelial ovarian carcinoma were 5 % , 14.3 % , 40.6 % and 67.5 % , respectively .
There was significant difference between borderline group and malignant group ( P0.05 ) .
The difference was significant between the normal group and the borderline group , the benign group and the borderline group ( P0.05 ) .
But there was no significant difference between the normal group and benign group ( P0.05 ) .

2 . In the patients with epithelial ovarian cancer , the positive rate of Cyclin E protein in the tumor tissues of high and high school differentiated patients was lower than that in the low differentiation group ( p < 0.05 ) .
