缺氧誘導(dǎo)因子-1α與缺氧誘導(dǎo)因子-2α蛋白和mRNA在宮頸癌中的表達(dá)及其臨床意義

【摘要】:目的探討缺氧誘導(dǎo)因子(HIF)-1α和HIF-2α在宮頸癌中的表達(dá)及其與患者臨床病理特征的相關(guān)性。方法用免疫組化法、蛋白印跡(Western blot)和實(shí)時(shí)定量聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)技術(shù)檢測(cè)30例宮頸癌患者(試驗(yàn)組)癌組織及30例宮頸良性病變患者(對(duì)照組)組織中HIF-1α和HIF-2α蛋白及mRNA的表達(dá),并分析其與宮頸癌臨床病理特征的相關(guān)性。結(jié)果 HIF-1αmRNA,HIF-2αmRNA在試驗(yàn)組中相對(duì)表達(dá)量為對(duì)照組的(2.06±0.68),(1.62±0.43),差異均有統(tǒng)計(jì)學(xué)意義(均P0.01)。免疫組化結(jié)果顯示,試驗(yàn)組中HIF-1α和HIF-2α的陽性表達(dá)率分別為63.33%和70.00%,對(duì)照組分別為16.67%和13.33%,差異均有統(tǒng)計(jì)學(xué)意義(均P0.01)。Western blot提示,試驗(yàn)組HIF-1α和HIF-2α蛋白表達(dá)量分別為(0.71±0.01)和(0.71±0.03),對(duì)照組分別為(0.53±0.03)和(0.56±0.05),差異均有統(tǒng)計(jì)學(xué)意義(均P0.05)。HIF-1α表達(dá)與淋巴結(jié)轉(zhuǎn)移及放療前貧血相關(guān)(均P0.05),HIF-2α表達(dá)與放療前貧血相關(guān)(P0.05)。結(jié)論試驗(yàn)組中,HIF-1α、HIF-2α在mRNA及蛋白質(zhì)水平均出現(xiàn)明顯升高,在宮頸癌的發(fā)展過程中可能起促癌作用;且兩者的表達(dá)呈同向變化,提示他們可能通過不同機(jī)制起著相同或相仿的作用。
[Abstract]:Objective to investigate the expression of HIF-1 偽 and HIF-2 偽 in cervical carcinoma and their correlation with clinicopathological features. Methods the expressions of HIF-1 偽 and HIF-2 偽 and mRNA were detected by immunohistochemistry, Western blot and real-time quantitative polymerase chain reaction (RT-PCR) in 30 patients with cervical cancer (trial group) and 30 patients with benign cervical lesions (control group). The relationship between the clinicopathological features of cervical cancer and its clinical features was analyzed. Results the relative expression of HIF-1 偽 mRNA-HIF-2 偽 mRNA in the experimental group was 2.06 鹵0.68, 1.62 鹵0.43, and the difference was statistically significant (all P 0.01). The results of immunohistochemistry showed that the positive rates of HIF-1 偽 and HIF-2 偽 were 63.33% and 70.005% in the experimental group and 16.67% and 13.33% in the control group, respectively. The expressions of HIF-1 偽 and HIF-2 偽 in the experimental group were 0.71 鹵0.01 and 0.71 鹵0.03, respectively, and those in the control group were 0.53 鹵0.03 and 0.56 鹵0.05, respectively. The difference was statistically significant (all the expressions of P0.05).HIF-1 偽 were correlated with lymph node metastasis and anemia before radiotherapy (all P0.05HIF-2 偽 expression was associated with anemia before radiotherapy). Conclusion in the experimental group, the level of HIF-1 偽 and HIF-2 偽 increased significantly in mRNA and protein levels, which may play a promoting role in the development of cervical cancer, and the expression of HIF-1 偽 and HIF-2 偽 may play the same or similar role in the development of cervical cancer, suggesting that they may play the same or similar role through different mechanisms.
