MicroRNA-182在宮頸癌中的表達及對腫瘤細胞增殖、侵襲及遷移的影響

【摘要】:目的 探討Micro RNA(mi R-182)在宮頸癌組織、宮頸上皮內(nèi)瘤變組織(CIN)和正常宮頸組織中的表達,及其對宮頸癌細胞增殖、侵襲和遷移能力的影響。方法 應用實時聚合酶鏈反應(real-time PCR)技術方法檢測78例宮頸癌組織、30例CIN組織及20例正常宮頸組織中mi R-182的表達,并分析其與宮頸癌臨床病理特征之間的關系。在宮頸癌細胞株Ca Ski中,應用細胞轉染技術上調(diào)mi R-182的表達,應用Cell Counting Kit-8(CCK-8)試劑盒檢測細胞增殖能力的變化,Transwell實驗檢測細胞侵襲遷移能力的變化。結果 Realtime PCR結果顯示mi R-182在宮頸癌組織、宮頸上皮內(nèi)瘤變(CIN)組織與正常宮頸組織比較,差異有統(tǒng)計學意義P0.05㖞;兩兩比較,mi R-182在宮頸癌組織和CIN組織中的表達較正常宮頸組織降低P0.05㖞,mi R-182在宮頸癌組織與CIN組織比較,差異無統(tǒng)計學意義(P0.05),宮頸癌組織中mi R-182的表達與分化程度、淋巴轉移密切相關,而與年齡、腫瘤直徑、浸潤深度及臨床分期等無關(P0.05)。在人宮頸癌上皮細胞(Ca Ski)中,上調(diào)mi R-182表達能夠抑制腫瘤細胞的侵襲和遷移P0.05㖞,但不改變細胞的增殖能力。結論 mi R-182在宮頸癌組織中低表達,mi R-182的表達水平與分化程度和淋巴結轉移有關,上調(diào)mi R-182的表達能夠抑制宮頸癌Ca Ski細胞的遷移及侵襲。
[Abstract]:Objective to investigate the expression of Micro RNA (MI R-182) in cervical cancer tissue, cervical intraepithelial neoplasia (CIN) and normal cervical tissue, and its effect on the proliferation, invasion and migration of cervical cancer cells. Methods the real-time polymerase chain reaction (real-time PCR) technique was used to detect 78 cervical cancer tissues, 30 CIN tissues and 20 normal cases. The expression of MI R-182 in cervical tissue and its relationship with the clinicopathological features of cervical cancer. In Ca Ski of cervical cancer cell line, the expression of MI R-182 was up-regulated by cell transfection technique, the changes of cell proliferation ability were detected by Cell Counting Kit-8 (CCK-8) kit, and the changes of cell invasion and migration ability were detected by Transwell experiment. Results Realtime PCR results showed that MI R-182 was compared with normal cervical tissue in cervical cancer tissue, cervical intraepithelial neoplasia (CIN) tissue and normal cervical tissue, the difference was statistically significant? P0.05? (22) the expression of MI R-182 in cervical cancer tissue and CIN tissue was lower than normal cervical tissue? P0.05? Mi R-182 in cervical cancer tissue compared with the tissue of cervical cancer. There is no statistical significance (P0.05). The expression of MI R-182 in cervical cancer tissues is closely related to the degree of differentiation and lymphatic metastasis, but not related to age, tumor diameter, depth of invasion and clinical staging (P0.05). In human cervical cancer epithelial cells (Ca Ski), up regulation of MI R-182 can inhibit the invasion and migration of tumor cells? P0.05? Conclusion the expression of MI R-182 in cervical cancer tissues is low, and the expression level of MI R-182 is related to the degree of differentiation and lymph node metastasis. The expression of MI R-182 can inhibit the migration and invasion of Ca Ski cells in cervical cancer.
