【摘要】:目的研究宮內(nèi)發(fā)育遲緩(IUGR)大鼠骨骼肌胰島素刺激下葡萄糖轉(zhuǎn)運蛋白4(GLUT4)表達和轉(zhuǎn)位的變化,探討IUGR個體發(fā)生胰島素抵抗的分子機制。方法采用孕期全程低蛋白飲食法建立大鼠IUGR模型,檢測18月齡IUGR子鼠骨骼肌胰島素刺激下的2-D-3H葡萄糖的攝取,同時采用實時熒光定量RT-PCR和Western blot方法檢測胰島素受體(IR)、胰島素受體底物1(IRS-1)及葡萄糖轉(zhuǎn)運蛋白4(GLUT4)的m RNA和蛋白表達;用Western blot方法分別檢測胰島素刺激前后骨骼肌總/膜GLUT4的蛋白表達變化。結(jié)果 1與對照組相比,18月齡IUGR組大鼠骨骼肌胰島素刺激后葡萄糖攝取率顯著降低(P=0.035);2IR和IRS-1 m RNA和蛋白表達在兩組大鼠骨骼肌中差異無統(tǒng)計學意義(P0.05),但GLUT4 m RNA表達在IUGR組大鼠骨骼肌中顯著下降(P=0.031);3基礎(chǔ)狀態(tài)時,兩組大鼠骨骼肌GLUT4總蛋白表達無明顯差異(P=0.967);IUGR組GLUT4膜蛋白表達略降低,但未及統(tǒng)計學意義(P=0.072);經(jīng)胰島素刺激后,兩組大鼠骨骼肌與各自基礎(chǔ)狀態(tài)相比,GLUT4總蛋白變化不明顯,GLUT4膜蛋白表達均較各自的基礎(chǔ)狀態(tài)顯著增加,但IUGR大鼠骨骼肌GLUT4膜蛋白的上升程度顯著低于對照組(P=0.001)。結(jié)論 IUGR老齡大鼠骨骼肌中GLUT4 m RNA表達降低以及胰島素刺激下的GLUT4轉(zhuǎn)位下降,可能降低對胰島素的反應性,從而減少葡萄糖攝取,促進胰島素抵抗發(fā)生。
[Abstract]:Objective to investigate the expression and translocation of glucose transporter 4 (GLUT4) in skeletal muscle of rats with intrauterine growth retardation (IUGR), and to explore the molecular mechanism of insulin resistance in IUGR individuals. Methods the IUGR model of rats was established by low-protein diet during pregnancy, and the 2-D-3H glucose uptake in skeletal muscle of 18 month old IUGR offspring was measured by insulin stimulation. Meanwhile, real-time quantitative RT-PCR and Western blot were used to detect the expression of m RNA and protein in insulin receptor irons, insulin receptor substrates 1 (IRS-1) and glucose transporter 4 (GLUT4). The protein expression of total / membrane GLUT4 in skeletal muscle was detected by Western blot before and after insulin stimulation. Results 1 compared with the control group, the glucose uptake rate of skeletal muscle in 18-month-old IUGR group was significantly lower than that in the control group after insulin stimulation. There was no significant difference in the expression of P0.035 IR and IRS-1 m RNA and protein between the two groups, but the expression of GLUT4 m RNA in the skeletal muscle of the two groups was not significantly different from that in the control group. In the IUGR group, the skeletal muscle decreased significantly in the basic state of P0. 031. There was no significant difference between the two groups in the expression of total GLUT4 protein in skeletal muscle. The expression of GLUT4 membrane protein in the group of GLUT4 0.967 was slightly decreased, but there was no significant difference between the two groups in the expression of GLUT4 membrane protein. The expression of GLUT4 membrane protein in skeletal muscle of the two groups was significantly higher than that of the control group, but the increase of GLUT4 membrane protein in the skeletal muscle of IUGR rats was significantly lower than that in the control group (P 0.001). Conclusion the decrease of GLUT4 m RNA expression in skeletal muscle of aged IUGR rats and the decrease of GLUT4 translocation induced by insulin may decrease the response to insulin, thereby reducing glucose uptake and promoting insulin resistance.
