【摘要】：目的探討低分子量酪氨酸磷酸酶(LMW-PTP)在子宮頸鱗癌中的表達(dá)及其臨床意義。方法采用RT-PCR及免疫組化法分析HaCaT、C33A、SiHa和CaSki細(xì)胞系中LMWPTP的表達(dá)情況。免疫組織化學(xué)法檢測(cè)正常宮頸患者19例,宮頸上皮內(nèi)瘤變(CIN)Ⅰ期患者18例、CINⅡ~Ⅲ患者46例和宮頸鱗癌患者76例的組織切片中LMW-PTP的表達(dá)。結(jié)果 LMW-PTP mRNA及蛋白在C33A、SiHa及CaSki宮頸癌細(xì)胞系中表達(dá)明顯高于人永生化正常表皮細(xì)胞系HaCaT;臨床組織樣本中,正常宮頸上皮LMW-PTP幾乎不表達(dá),在正常宮頸上皮到CIN繼而發(fā)展成宮頸癌的進(jìn)展過程中LMW-PTP的表達(dá)逐漸升高,差異有統(tǒng)計(jì)學(xué)意義(P0.05);低分化宮頸癌組織中LMW-PTP表達(dá)高于中分化組織(P=0.025),中分化宮頸癌組織中LMW-PTP的表達(dá)高于高分化組織(P=0.045);宮頸癌組織中LMW-PTP蛋白的表達(dá)與組織學(xué)分級(jí)程度和是否有淋巴結(jié)轉(zhuǎn)移密切相關(guān)(P0.05)。結(jié)論LMW-PTP在宮頸癌的進(jìn)展過程中起重要作用,且與組織分化程度和是否有淋巴轉(zhuǎn)移密切相關(guān),提示LMW-PTP的表達(dá)有助于判斷宮頸癌患者的預(yù)后。
