【摘要】：目的:探討子宮內(nèi)膜腺癌組織中胰島素樣生長因子結(jié)合蛋白相關(guān)蛋白1(IGFBP-r P1)基因的甲基化狀態(tài)及其與IGFBP-r P1 mRNA、蛋白表達(dá)之間的關(guān)系。方法:采用甲基化特異性PCR檢測45例子宮內(nèi)膜腺癌、30例子宮內(nèi)膜單純性增生和30例子宮內(nèi)膜不典型增生組織中IGFBP-r P1基因啟動子區(qū)和第一外顯子區(qū)Cp G島的甲基化狀態(tài);采用實時熒光定量PCR和免疫組化SP法分別檢測上述3種組織中IGFBP-r P1 mRNA和蛋白的表達(dá)。結(jié)果:子宮內(nèi)膜腺癌組織中IGFBP-r P1基因在啟動子區(qū)的甲基化率高于其在第一外顯子區(qū)的甲基化率(χ2=6.429,P=0.011)。子宮內(nèi)膜腺癌組織中IGFBP-r P1基因在啟動子區(qū)的甲基化率低于子宮內(nèi)膜不典型增生組織和單純性增生組織(F=14.659,P=0.001)。子宮內(nèi)膜腺癌組織中IGFBP-r P1 mRNA和蛋白的表達(dá)均高于子宮內(nèi)膜不典型增生組織和單純性增生組織(F=8.619、χ2=23.611,P均0.05);在啟動子區(qū)發(fā)生IGFBP-r P1基因甲基化的子宮內(nèi)膜腺癌組織中,其mRNA的表達(dá)水平低于未甲基化組(t=4.758,P=0.001),其蛋白的表達(dá)與甲基化狀態(tài)呈負(fù)關(guān)聯(lián)(rP=-0.625,P0.001)。結(jié)論:子宮內(nèi)膜腺癌組織中IGFBP-r P1基因甲基化主要發(fā)生在啟動子區(qū),且呈低甲基化狀態(tài);IGFBP-r P1啟動子區(qū)的低甲基化狀態(tài)可能是IGFBP-r P1基因在子宮內(nèi)膜腺癌組織中高表達(dá)的調(diào)控機(jī)制之一。
[Abstract]:Aim: to investigate the methylation status of insulin-like growth factor binding protein-associated protein (1(IGFBP-r P1) gene in endometrial adenocarcinoma and its relationship with the expression of IGFBP-r P1 mRNAs and proteins.Methods: methylation-specific PCR was used to detect the methylation status of CP G islands in the promoter region and the first exon region of IGFBP-r P1 gene in 30 cases of endometrial simple hyperplasia and 30 cases of endometrial atypical hyperplasia in 45 cases of endometrial adenocarcinoma and 30 cases of endometrial atypical hyperplasia.The expression of IGFBP-r P1 mRNA and protein in these three tissues was detected by real-time fluorescence quantitative PCR and immunohistochemical SP method.Results: the methylation rate of IGFBP-r P1 gene in the promoter region of endometrial adenocarcinoma was higher than that in the first exon (蠂 2 6.429).The methylation rate of IGFBP-r P1 gene in endometrial adenocarcinoma was lower than that in atypical endometrial hyperplasia and simple hyperplasia.The expression of IGFBP-r P1 mRNA and protein in endometrial adenocarcinoma was higher than that in atypical endometrial hyperplasia and simple hyperplasia (P < 0.05).The expression level of mRNA was lower than that of unmethylated group, and the protein expression was negatively correlated with methylation status.Conclusion: the methylation of IGFBP-r P1 gene mainly occurs in the promoter region of endometrial adenocarcinoma.The hypomethylation of IGFBP-r P1 promoter may be one of the regulatory mechanisms of IGFBP-r P1 gene overexpression in endometrial adenocarcinoma.
