【摘要】:目的本研究意在探討SIX1蛋白在人宮頸癌組織中的表達(dá)情況及與宮頸癌生物學(xué)特性的關(guān)系。 方法所選實(shí)驗(yàn)標(biāo)本例數(shù)共204例,組織芯片包括107例宮頸鱗癌,16例宮頸腺癌,另有32例正常宮頸、49例宮頸原位癌。應(yīng)用免疫熒光染色法觀察SIX1基因在HELA細(xì)胞系中的細(xì)胞定位情況,采用免疫組化SP法檢測SIX1蛋白在各種宮頸組織中的表達(dá)情況,并分析SIX1蛋白的表達(dá)與宮頸癌臨床病理學(xué)特征的關(guān)聯(lián)。 結(jié)果1:免疫熒光染色發(fā)現(xiàn):SIX1蛋白在宮頸癌HeLa細(xì)胞系中主要定位于細(xì)胞漿和核仁。 2:從正常宮頸上皮到CIN原位癌再到浸潤癌,SIX1蛋白陽性表達(dá)量有顯著升高趨勢。與正常宮頸上皮組織相比,宮頸癌組織中SIX1蛋白陽性率及陽性強(qiáng)度明顯升高,差異有統(tǒng)計(jì)學(xué)意義(P0.001)。 3:宮頸癌中SIX1蛋白的表達(dá)與腫瘤大小、淋巴轉(zhuǎn)移、血液轉(zhuǎn)移有關(guān),有統(tǒng)計(jì)學(xué)差異(P0.05或P0.01),而與年齡、腫瘤分期以及腫瘤分化程度無關(guān)(P0.05)。 結(jié)論1:SIXl蛋白的過表達(dá),可能參與了宮頸癌的發(fā)生發(fā)展過程。2.SIX1蛋白的表達(dá)與宮頸癌的生物學(xué)特性--侵襲和轉(zhuǎn)移性行為有關(guān)。在臨床工作中為宮頸癌的侵襲與轉(zhuǎn)移提供了可能的分子標(biāo)志物。
[Abstract]:Objective to investigate the expression of SIX1 protein in human cervical carcinoma and its relationship with the biological characteristics of cervical cancer.Methods A total of 204 samples were selected, including 107 cases of squamous cell carcinoma of the cervix and 16 cases of adenocarcinoma of the cervix, and there were 32 cases of normal cervical carcinoma in situ.The localization of SIX1 gene in HELA cell line was observed by immunofluorescence staining, and the expression of SIX1 protein in various cervical tissues was detected by immunohistochemical SP method.The relationship between the expression of SIX1 protein and clinicopathological features of cervical cancer was analyzed.Results 1. It was found by immunofluorescence staining that the protein was mainly located in cytoplasm and nucleoli in cervical cancer HeLa cell line.2: the positive expression of SIX 1 protein increased significantly from normal cervical epithelium to CIN carcinoma in situ and then to invasive carcinoma.Compared with normal cervical epithelium, the positive rate and intensity of SIX1 protein in cervical carcinoma tissue were significantly higher than those in normal cervical epithelium tissue (P 0.001).3. The expression of SIX1 protein in cervical cancer was related to tumor size, lymphatic metastasis and blood metastasis, there was significant difference (P0.05 or P0.01), but it was not related to age, tumor stage and tumor differentiation.Conclusion the overexpression of 1:SIXl protein may be involved in the genesis and development of cervical cancer. 2. The expression of SIX1 protein may be related to the biological characteristics of cervical cancer, invasion and metastasis.It provides a possible molecular marker for invasion and metastasis of cervical cancer.
