XPG基因遺傳變異與上皮性卵巢癌鉑類化療預(yù)后相關(guān)性研究

【摘要】:目的探討XPG基因單核苷酸多態(tài)性(SNPs)與上皮性卵巢癌鉑類化療預(yù)后的關(guān)系。方法通過(guò)Haploview軟件篩選出XPG基因9個(gè)標(biāo)簽SNPs(tag SNPs)。連接酶檢測(cè)反應(yīng)技術(shù)檢測(cè)2002年3月至2009年12月在河北醫(yī)科大學(xué)第四醫(yī)院進(jìn)行手術(shù)治療的239例上皮性卵巢癌患者外周血DNA XPG基因9個(gè)tag SNPs的基因型頻率分布情況。采用生存分析評(píng)估XPG基因SNPs與上皮性卵巢癌患者臨床預(yù)后的關(guān)系。患者臨床預(yù)后的判定指標(biāo)包括患者對(duì)鉑類化療反應(yīng)(TR)、疾病無(wú)進(jìn)展生存期(PFS)和總生存期(OS)。結(jié)果統(tǒng)計(jì)學(xué)分析顯示:XPG基因rs1047768C/T和rs2296147C/T多態(tài)可能與確診時(shí)年齡≥50歲的卵巢癌患者臨床預(yù)后相關(guān)。即與rs1047768 T/T基因型相比,rs1047768C等位基因降低了患者鉑類抵抗、疾病復(fù)發(fā)和死亡的風(fēng)險(xiǎn)(TR:OR=0.49,95%CI 0.24~0.98;PFS:HR=0.57,95%CI 0.38~0.87;OS:HR=0.56,95%CI 0.34~0.94);與rs2296147 T/T基因型相比,rs2296147C等位基因降低上皮性卵巢癌患者疾病復(fù)發(fā)和死亡的風(fēng)險(xiǎn)(PFS:HR=0.63,95%CI 0.41~0.98;OS:HR=0.50,95%CI 0.28~0.87)。結(jié)論 XPG rs1047768C/T和rs2296147C/T多態(tài)可能是確診時(shí)年齡≥50歲卵巢癌患者臨床預(yù)后的分子標(biāo)志物。
[Abstract]:Objective to investigate the relationship between XPG gene single nucleotide polymorphisms (SNPs) and the prognosis of platinum chemotherapy in epithelial ovarian cancer.Methods 9 XPG gene tags SNPs(tag SNPs were screened by Haploview software.The genotypes of DNA XPG gene 9 tag SNPs in peripheral blood of 239 patients with epithelial ovarian cancer treated in the fourth Hospital of Hebei Medical University from March 2002 to December 2009 were detected by ligase assay.Survival analysis was used to evaluate the relationship between XPG gene SNPs and clinical prognosis in patients with epithelial ovarian cancer.The clinical prognostic criteria included the response of patients to platinum chemotherapy, the progressive survival time (PFS) and the total survival time (TSA).Results the statistical analysis showed that the rs1047768C/T and rs2296147C/T polymorphisms of the 1: XPG gene might be related to the clinical prognosis of ovarian cancer patients aged more than 50 years at the time of diagnosis.Conclusion Polymorphism of XPG rs1047768C/T and rs2296147C/T may be molecular markers for clinical prognosis of ovarian cancer patients aged 鈮,
