CTLA-4基因多態(tài)性與宮頸癌易感性的關(guān)系

【摘要】:目的探討細(xì)胞毒T淋巴細(xì)胞相關(guān)抗原(CTLA)-4基因單核苷酸多態(tài)性(SNPs)與宮頸癌發(fā)生的關(guān)系。方法 350例宮頸癌患者為病例組,350例健康女性為對照組,抽取全血基因組DNA,直接測序法檢測CTLA-4-318 T/C、CT60 G/A單核苷酸多態(tài)性,采用MTT法檢測不同基因型的外周血單核細(xì)胞增殖能力,酶聯(lián)免疫吸附(ELISA)法檢測不同基因型患者血清轉(zhuǎn)化生長因子(TGF)-β、白細(xì)胞介素(IL)-2、IL-4表達(dá)水平。結(jié)果宮頸癌病例組CTLA-4-318位點(diǎn)等位基因C的概率低于對照組,等位基因T概率高于對照組,CC基因型比例低于對照組,TT基因型比例高于對照組(均P0.05)。病例組CTLA-4 CT60位點(diǎn)等位基因A的概率低于對照組,等位基因G的概率高于對照組,AA基因型比例低于對照組,GG基因型比例高于對照組(均P0.05)。CTLA-4-318 CC基因型T細(xì)胞增殖率高于TT基因型,TGF-β水平低于TT基因型,IL-2、IL-4水平高于TT基因型(均P0.05)。CTLA-4 CT60 AA基因型TGF-β水平低于GG基因型,IL-2、IL-4水平高于GG基因型(P0.05)。結(jié)論 CTLA-4-318 T/C、CT60 G/A基因多態(tài)性在宮頸癌的發(fā)生發(fā)展過程中起到重要作用,CTLA-4-318 TT基因型及CT60 GG基因型可增加宮頸癌的發(fā)病風(fēng)險(xiǎn),其機(jī)制可能與調(diào)節(jié)T細(xì)胞的增殖及調(diào)節(jié)細(xì)胞因子的分泌有關(guān)。
[Abstract]:Objective to investigate the relationship between cytotoxic T lymphocyte associated antigen (CTLA-4) single nucleotide polymorphisms (SNPs) and cervical carcinogenesis. The genomic DNA of whole blood was extracted, and the single nucleotide polymorphisms of CTLA-4-318 T / CT60 G / A were detected by direct sequencing. The proliferative ability of peripheral blood monocytes with different genotypes was detected by MTT method. The levels of TGF- 尾 and IL-4 in serum of patients with different genotypes were detected by Elisa. Results the probability of allele C at CTLA-4-318 locus in cervical cancer patients was lower than that in control group. The probability of allele T was higher than that of control group (P < 0.05). The probability of allele A at CTLA-4 CT60 locus in case group was lower than that in control group. The probability of allele G was higher than that of the control group. The percentage of AA genotype was lower than that of the control group. The percentage of T cell proliferation of P0.05).CTLA-4-318 CC genotype was higher than that of the control group. The level of TGF- 尾 was lower than that of TGF- 尾 of TT genotype and the level of TGF- 尾 was higher than that of TT genotype. The level of TGF- 尾 of P0.05).CTLA-4 CT60 AA genotype was lower than that of GG genotype (P 0.05). Conclusion the polymorphism of CTLA-4-318 T / CfCT60 G / A gene plays an important role in the occurrence and development of cervical cancer. CTLA-4-318TT genotype and CT60 GG genotype may play an important role in the occurrence and development of cervical cancer. Increase the risk of cervical cancer, The mechanism may be related to the regulation of T cell proliferation and cytokine secretion.
