【摘要】：目的探討上皮性卵巢癌(EOC)中八聚體結(jié)合轉(zhuǎn)錄因子4(OCT4)、Notch1及DLL4蛋白表達之間的關(guān)系及其功能意義。方法收集207例EOC術(shù)后標本和65例卵巢良性上皮性腫瘤標本,應用免疫組化學法檢測EOC和卵巢良性上皮性腫瘤組織中OCT4、Notch1以及DLL4蛋白的表達情況。結(jié)果 EOC組織和卵巢良性上皮性腫瘤組織中,OCT4、Notch1和DLL4蛋白的陽性表達率分別為60.0%、61.8%、60.9%和9.2%、6.2%、0,差異均有統(tǒng)計學意義(P0.05);OCT4、Notch1和DLL4的表達與EOC的組織學分化、FIGO分期、盆腔淋巴結(jié)轉(zhuǎn)移有關(guān)(P0.05);DLL4蛋白的表達分別與OCT4和Notch1的表達呈正相關(guān)關(guān)系(r值分別為0.758和0.704,P均0.001),同時,OCT4與Notch1蛋白的表達亦呈正相關(guān)(r=0.645,P0.001)。Kaplan-Meier生存分析表明,OCT4、Notch1和DLL4蛋白的過表達均與患者的生存率有關(guān),3種蛋白表達陽性組患者生存率均明顯低于蛋白表達陰性組患者(P0.05)。多因素分析:OCT4、DLL4蛋白的表達和FIGO分期是影響EOC根治術(shù)后患者預后的獨立因素(P0.05)。結(jié)論OCT4、Notch1及DLL4的過表達與EOC的組織學分化程度、轉(zhuǎn)移和預后等因素密切相關(guān),聯(lián)合檢測這些指標可能對于EOC患者的病情進展和預后判斷給予重要提示。
[Abstract]:Objective to investigate the relationship between the expression of octamer binding transcription factor 4OCT4tch1 and DLL4 protein in epithelial ovarian carcinoma (EOC). Methods 207 cases of EOC postoperative specimens and 65 cases of ovarian benign epithelial tumors were collected. The expression of OCT4ntch1 and DLL4 protein in EOC and ovarian benign epithelial tumors were detected by immunohistochemical method. Results the positive rates of OCT4Notch1 and DLL4 in EOC and ovarian benign epithelial tumors were 60.0% and 60.9%, respectively. There were significant differences in the expression of OCT4Notch1 and DLL4 and the histological differentiation of EOC in Figo stage. The expression of DLL4 protein in pelvic lymph node metastasis was positively correlated with the expression of OCT4 and Notch1. The r values were 0.758 and 0.704, respectively. The expression of OCT4 and Notch1 protein was also positively correlated with the expression of Notch1 protein. The survival analysis showed that OCT4Notch1 and DLL4 protein were overexpressed. The survival rate of the patients with positive expression of three proteins was significantly lower than that of the patients with negative protein expression (P 0.05). Multivariate analysis showed that the expression of DLL4 protein and the FIGO stage were independent of the prognosis of patients after EOC radical operation. Conclusion the overexpression of OCT4, Notch1 and DLL4 and the histological differentiation of EOC may be related to the expression of OCT4, Notch1 and DLL4. The factors such as metastasis and prognosis are closely related. The combined detection of these indicators may be an important indication for the progression and prognosis of EOC patients.
