【摘要】：目的 :探討糖原合成酶激酶3β(glycogen synthase kinase,GSK-3β)對卵巢癌順鉑耐藥性的影響及機(jī)制。方法 :運(yùn)用氯化鋰(lithium chloride,Li Cl)增加失活性GSK-3β即p-GSK-3β表達(dá),MTT法篩選合適的Li Cl濃度和作用時(shí)間并檢測順鉑的半數(shù)抑制濃度,流式細(xì)胞儀檢測p-GSK-3β(ser9)的表達(dá)量,免疫熒光觀察其細(xì)胞內(nèi)定位情況。結(jié)果:據(jù)MTT結(jié)果選擇Li Cl 20 mmol/L處理6 h,處理后24、48、72 h,SKOV3細(xì)胞順鉑的半數(shù)抑制濃度從處理前的(20.37±0.56)、(7.79±0.72)、(2.8±0.32)μg/ml增加至(27.57±0.81)、(12.15±1.00)、(5.84±0.64)μg/ml,差異均有統(tǒng)計(jì)學(xué)意義。流式結(jié)果顯示,Li Cl處理后SKOV3細(xì)胞內(nèi)p-GSK-3β(ser9)的平均熒光強(qiáng)度由處理前的661.67±37.63增加到1 191.33±30.37,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。免疫熒光結(jié)果提示,Li Cl處理后SKOV3細(xì)胞核內(nèi)p-GSK-3β(ser9)表達(dá)上調(diào)。結(jié)論:Li Cl預(yù)處理后卵巢癌細(xì)胞順鉑耐藥性的增強(qiáng)可能與GSK-3β失活性磷酸化水平增加,尤其是與核內(nèi)水平上調(diào)有關(guān)。
[Abstract]:Objective: to investigate the effect and mechanism of glycogen synthase kinase 3 尾 -glycogen synthase kinase- GSK-3 尾 on cisplatin resistance in ovarian cancer. Methods: Lithium chloridede LiCl) was used to increase the inactivation of GSK-3 尾, or p-GSK-3 尾 expression. The half inhibitory concentration of cisplatin was detected. Flow cytometry was used to detect the expression of p-GSK-3 尾 -ser9. Results: the intracellular localization of SKOV3 cells was observed by immunofluorescence. Results: according to the MTT results, the median inhibitory concentration of cisplatin in SKOV3 cells was increased from 7.79 鹵0.72 鹵0.72 渭 g/ml to 27.57 鹵0.81 鹵1.00 鹵5.84 渭 g / ml, respectively. The results showed that the average fluorescence intensity of SKOV3 cells increased from 661.67 鹵37.63 to 1 191.33 鹵30.37 after LiCl treatment, and the difference was statistically significant (P 0.05). The immunofluorescence results suggested that the expression of p-GSK-3 尾 ser9 in SKOV3 cells was up-regulated after LiCl treatment. The increase of cisplatin resistance may be associated with the increase of GSK-3 尾 deactivated phosphorylation. In particular, it is related to the upregulation of nuclear level.
