胡桃醌對(duì)人宮頸癌HeLa細(xì)胞侵襲與遷移的影響

【摘要】:目的觀察胡桃醌對(duì)HeLa細(xì)胞侵襲及遷移的抑制作用,并探討其可能的作用機(jī)制。方法常規(guī)體外培養(yǎng)人HeLa細(xì)胞,加入10、20、50、100μmol/L濃度的胡桃醌,繼續(xù)培養(yǎng)24 h。通過倒置顯微鏡觀察細(xì)胞形態(tài)學(xué)變化,采用MTT法檢測(cè)細(xì)胞增殖情況,細(xì)胞劃痕實(shí)驗(yàn)檢測(cè)胡桃醌對(duì)細(xì)胞遷移能力的影響,用Transwell小室法檢測(cè)細(xì)胞侵襲能力的影響。Western blotting檢測(cè)基質(zhì)金屬蛋白酶(MMP)-2、MMP-9蛋白的表達(dá)。結(jié)果細(xì)胞形態(tài)學(xué)觀察顯示,不同濃度胡桃醌均可導(dǎo)致細(xì)胞形態(tài)學(xué)發(fā)生改變;MTT檢測(cè)表明,胡桃醌對(duì)HeLa細(xì)胞的生長(zhǎng)有明顯抑制作用,且抑制作用隨藥物濃度的增加而增強(qiáng)(P0.05或P0.01);劃痕實(shí)驗(yàn)結(jié)果顯示,隨胡桃醌藥物濃度的增加,細(xì)胞的平面運(yùn)動(dòng)能力明顯下降;Transwell侵襲小室實(shí)驗(yàn)結(jié)果顯示,胡桃醌對(duì)HeLa細(xì)胞體外的侵襲力具有很強(qiáng)的抑制作用;Western blotting結(jié)果表明,胡桃醌給藥能夠抑制HeLa細(xì)胞MMP-2、MMP-9蛋白的表達(dá)。結(jié)論胡桃醌能夠抑制HeLa細(xì)胞的增殖并抑制細(xì)胞侵襲,其機(jī)制可能與抑制MMP-2、MMP-9的表達(dá)有關(guān)。
[Abstract]:Objective to observe the inhibitory effect of walnut quinone on the invasion and migration of HeLa cells and to explore its possible mechanism. The morphological changes of cells were observed by inverted microscope, cell proliferation was detected by MTT assay, and the effect of walnut quinone on cell migration was detected by cell scratch assay. Transwell chamber assay was used to detect the cell invasion ability. Western blotting was used to detect the expression of matrix metalloproteinase MMP-2MMP-2MMP-9 protein. The growth of HeLa cells was inhibited by nutanone, and the inhibitory effect increased with the increase of drug concentration (P 0.05 or P 0.01), and the scratch test showed that the inhibitory effect increased with the increase of drug concentration of pectin. The results of transwell invasive chamber experiments showed that walnut quinone had a strong inhibitory effect on the invasion of HeLa cells in vitro. Conclusion Nutanone can inhibit the proliferation and invasion of HeLa cells, and its mechanism may be related to the inhibition of MMP-2 MMP-9 expression.
