超聲介導(dǎo)正己酰羧甲基殼聚糖載藥納米液滴對(duì)卵巢癌細(xì)胞的抑制作用

【摘要】:目的評(píng)價(jià)采用正己酰羧甲基殼聚糖制備相變型載阿霉素(DOX)納米液滴的體外超聲顯像效果及抑制卵巢癌細(xì)胞生長(zhǎng)的作用。方法將羧甲基殼聚糖與正己酸酐發(fā)生酰化反應(yīng)合成正己酰羧甲基殼聚糖,采用聲振乳化法制備正己酰羧甲基殼聚糖載藥納米液滴,表征其表面形態(tài)、粒徑、電位;評(píng)價(jià)其體外超聲顯像效果,以紫外分光光度計(jì)測(cè)定藥物包封率,采用CCK-8法檢測(cè)卵巢癌細(xì)胞存活率,并進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果成功制備正己酰羧甲基殼聚糖載藥納米液滴,鏡下為大小均勻的球狀物,平均粒徑為(458.33±43.50)nm,包封率為(52.06±10.14)%。納米液滴可增強(qiáng)超聲顯影效果。超聲聯(lián)合載藥納米液滴組的卵巢癌細(xì)胞存活率[(62.54±3.60)%]低于DOX組[(75.55±7.21)%]、載藥納米液滴組[(76.18±4.94)%]和超聲組[(89.90±0.83)%],差異有統(tǒng)計(jì)學(xué)意義(P均0.05)。結(jié)論超聲聯(lián)合正己酰羧甲基殼聚糖載藥納米液滴可有效抑制卵巢癌細(xì)胞生長(zhǎng),在臨床診療中具有潛在應(yīng)用價(jià)值。
[Abstract]:Objective to evaluate the effect of ultrasonic imaging of phase change doxorubicin (DOX) nanoparticles prepared by hexanoyl carboxymethyl chitosan on ovarian cancer cell growth. Methods Carboxymethyl chitosan was acylated with hexanoic anhydride. Synthesis of n-hexyl carboxymethyl chitosan by reaction, Nano-droplets loaded with n-hexyl carboxymethyl chitosan were prepared by acoustical emulsification method, their surface morphology, particle size and potential were characterized, the effect of ultrasonic imaging in vitro was evaluated, and the encapsulation efficiency was measured by ultraviolet spectrophotometer. CCK-8 assay was used to detect the survival rate of ovarian cancer cells, and the results showed that n-hexyl carboxymethyl chitosan nanoparticles were successfully prepared. The average particle size was 458.33 鹵43.50 nm, the encapsulation efficiency was 52.06 鹵10.140.The sonographic effect was enhanced by nano-droplets. The survival rate of ovarian cancer cells in the combination of ultrasonic and drug-loaded nano-droplet group [62.54 鹵3.60%] was lower than that in DOX group [75.55 鹵7.21%], the drug loaded nano-droplet group [76.18 鹵4.94 渭%] and the ultrasound group [89.90 鹵0.833%], the difference was statistically significant. Conclusion Ultrasound combined with n-hexanoyl carboxymethyl chitosan nanoparticles can effectively inhibit the growth of ovarian cancer cells. It has potential application value in clinical diagnosis and treatment.
