Hemin對妊娠期高血壓疾病大鼠HO-1的表達、血壓及24h尿蛋白的影響

【摘要】:目的:(1)探討血紅素氧合酶-1與大鼠妊娠期高血壓疾病的相關性;(2)探討正鐵血紅素(Hemin)對妊娠期高血壓大鼠血紅素氧合酶.1(HO-1)的表達、血壓及24h尿蛋白的影響。 方法:(1)建模及干預:正常SD雌性大鼠30只與雄性大鼠1:1合籠,雌鼠陰道分泌物發(fā)現(xiàn)精子計為妊娠第0天,妊娠第12天測量孕鼠血壓并收集24h尿量。妊娠第14天將孕鼠隨機分為三組:L-NAME組(A)、L-NAME+Heminta (B)、正常妊娠組(C),A、B組予亞硝基左旋精氨酸甲酯(L-NAME)80mg/kg/d灌胃建立大鼠妊娠期高血壓模型,C組予等量生理鹽水灌胃。妊娠第16天再次測量各組孕鼠血壓,A、B組血壓升高達到高血壓診斷標準者納入實驗,繼續(xù)每日灌胃L-NAME80mg/kg/d,共7天,B組同時于妊娠第16天始給予Hemin30mg/kg腹腔注射4天。(2)標本采集及指標檢測:妊娠第12天、妊娠第16天、妊娠第20天測量各組孕鼠血壓并收集24h尿量,用自動生化分析儀測定24h尿蛋白;妊娠第20天收集尿量完畢后取孕鼠胎盤進行HE染色觀察胎盤形態(tài)學變化及免疫組織化學法檢測胎盤組織中HO.1的定位和表達;腹主動脈采血用ELISA方法檢測血清中HO.1的表達。 結(jié)果:(1)獲妊娠大鼠21只,隨機分為三組:L-NAME組(A)、 L-NAME+Hemin組(B)、正常妊娠組(C),每組7只;(2)血壓(mmHg):妊娠第16天A、B組各7只孕鼠血壓均升高達大鼠高血壓診斷標準,均納入實驗,A、B組孕鼠血壓均值均高于C組,且均較同組妊娠第12天的基礎血壓明顯升高,差異有統(tǒng)計學意義(P0.05),建模成功;妊娠第20天A、B組孕鼠血壓仍高于C組,B組血壓較A組下降且低于同組妊娠第16天血壓,差異均有統(tǒng)計學意義(P0.05)。(3)24h尿蛋白(mg/24h):妊娠第20天A、B組孕鼠24h尿蛋白較C組明顯升高,且均較同組妊娠第12天及妊娠第16天明顯升高,差異有統(tǒng)計學意義(P0.05);妊娠第20天B組較A組24h尿蛋白雖有下降趨勢,但差異無統(tǒng)計學意義(P0.05)。(4)血清中HO.1的表達:A、B、C組HO-1表達量依次增多,三組兩兩比較差異均有統(tǒng)計學意義(P0.05)。妊娠第20天孕鼠24h尿蛋白與血清中HO-1表達之間存在負相關關系,Pearson相關系數(shù)為.0.603。(5)免疫組織化學染色提示HO-1主要表達于胎盤海綿滋養(yǎng)層細胞,A、B、C組HO-1蛋白表達依次增多,三組兩兩比較差異均有統(tǒng)計學意義(P0.05)。 結(jié)論:(1)HO-1與大鼠妊娠期高血壓疾病具有相關性,可能為妊娠期高血壓疾病發(fā)病機制之一;(2)Hemin能夠上調(diào)HO.1的表達,降低血壓,但對24h尿蛋白的影響還需進一步探討,可能對妊娠期高血壓疾病具有一定的治療作用。
[Abstract]:Objective: (1) to explore the correlation between heme oxygenase -1 and hypertensive disorder complicating pregnancy. (2) to explore the effect of hemin (Hemin) on heme oxygenase.1 (HO-1) expression, blood pressure and 24h urinary protein in hypertensive rats.
Methods: (1) modeling and intervention: normal 30 female SD rats and male rats were mated female rats 1:1, vaginal secretions for sperm was found on day zeroth of pregnancy, Twelfth days of blood pressure measurement in pregnant rats and collect 24h urine pregnancy. Pregnancy fourteenth days pregnant rats were randomly divided into three groups: group L-NAME (A), L-NAME+Heminta (B), normal pregnancy group (C), A, B group were treated with L-NAME (L-NAME 80mg/kg/d) to establish the rat model of gastric lavage in hypertensive pregnancy, C group was treated with normal saline. The sixteenth day of pregnancy again measure blood pressure in pregnant rats of each group A, B group up to the diagnostic criteria of hypertension were included in the experiment, to gavage L-NAME80mg/kg/d, a total of 7 days, the B group on the sixteenth day of pregnancy was given intraperitoneal injection of Hemin30mg/kg for 4 days. (2) detection of specimen collection and index: Twelfth days of pregnancy, pregnancy sixteenth days, pregnancy twentieth days to measure the blood pressure of pregnant rats and collect group 24h urine volume, with Determination of 24h urine protein automatic biochemical analyzer; the expression and localization of the twentieth day of pregnancy to collect HO.1 urine volume after the placenta in pregnant rats by HE staining to observe the change of placental morphology and immunohistochemistry in the placenta tissue; the expression of abdominal aorta with ELISA method for detection of HO.1 in serum.
