【摘要】：目的構(gòu)建PIAS-1 siRNA干擾載體Plko.1、PIAS-1。方法設(shè)計(jì)以PIAS-1基因?yàn)榘袠?biāo)的siRNA序列,并克隆人載體Plko.1,用基因測(cè)序進(jìn)行重組克隆驗(yàn)證。結(jié)果 PCR檢測(cè)證實(shí)siRNA插入Plko.1質(zhì)粒,測(cè)序分析證實(shí)插入序列正確。結(jié)論成功構(gòu)建PIAS-1基因的siRNA載體,為研究PIAS-1基因?qū)m頸癌細(xì)胞增殖凋亡的影響提供了穩(wěn)定轉(zhuǎn)染的骨架載體。
[Abstract]:Objective to construct PIAS-1 siRNA interference vector Plko.1PIAS-1.Methods A siRNA sequence targeting PIAS-1 was designed and cloned into human vector Plko.1.Recombinant cloning was performed by gene sequencing. Results PCR analysis confirmed that siRNA was inserted into Plko.1 plasmid. Conclusion the siRNA vector of PIAS-1 gene was constructed successfully, which provides a stable transfection skeleton vector for studying the effect of PIAS-1 gene on the proliferation and apoptosis of cervical cancer cells.
