【摘要】：目的探討靶向沉默EZH2基因?qū)m頸癌細(xì)胞侵襲轉(zhuǎn)移的影響及其分子機(jī)制。方法采用化學(xué)合成siRNA瞬時(shí)轉(zhuǎn)染人宮頸癌C33A細(xì)胞,沉默EZH2基因的表達(dá)。通過細(xì)胞劃痕實(shí)驗(yàn)、Transwell小室侵襲實(shí)驗(yàn)、軟瓊脂克隆形成實(shí)驗(yàn),觀察沉默EZH2對宮頸癌細(xì)胞遷移、侵襲及體外成瘤能力的影響,Western blot法檢測MMP2的表達(dá)變化。結(jié)果 EZH2siRNA轉(zhuǎn)染C33A細(xì)胞后,細(xì)胞劃痕及Transwell小室侵襲實(shí)驗(yàn)結(jié)果均顯示宮頸癌細(xì)胞的遷移、侵襲能力明顯減低,差異具有統(tǒng)計(jì)學(xué)意義;軟瓊脂克隆形成實(shí)驗(yàn)表明,EZH2沉默后克隆形成數(shù)目明顯減少,差異具有統(tǒng)計(jì)學(xué)意義;Western blot結(jié)果顯示MMP2表達(dá)水平下調(diào)。結(jié)論沉默EZH2基因表達(dá)能顯著抑制宮頸癌C33A細(xì)胞的侵襲轉(zhuǎn)移能力,其作用機(jī)制可能通過下調(diào)MMP2來實(shí)現(xiàn)。
[Abstract]:Objective to investigate the effect of targeted silencing EZH2 gene on invasion and metastasis of cervical cancer cells and its molecular mechanism. Methods the expression of EZH2 gene was instantly transfected into human cervical cancer C33A cells by chemically synthesized siRNA. The effect of silencing EZH2 on the migration, invasion and tumorigenic ability of cervical cancer cells in vitro was observed. The expression of MMP2 was detected by Western blot. Results after transfection of EZH2siRNA into C33A cells, the expression of MMP2 was detected by Western blot. The results of cell scratch and Transwell chamber invasion showed that the migration and invasion ability of cervical cancer cells were significantly decreased, the difference was statistically significant, the results of soft Agar clone formation test showed that the number of colony formation was significantly decreased after EZH2 silencing. Conclusion silencing the expression of EZH2 gene can significantly inhibit the invasion and metastasis of cervical cancer C33A cells, and its mechanism may be achieved by down-regulation of MMP2.
