量子點(diǎn)免疫熒光原位雜交法檢測(cè)宮頸癌組織芯片中高危型HPV的感染狀況

【摘要】:目的研究宮頸癌及相關(guān)組織高危型HPV的感染狀況,并評(píng)價(jià)所用檢測(cè)方法的特異性和敏感性。方法回顧性采集124例石蠟包埋宮頸(癌)組織,包括80例宮頸鱗狀細(xì)胞癌,24例宮頸腺癌,以及20例因子宮肌瘤或子宮腺肌病等原因切除的正常宮頸組織或炎性宮頸組織。收集宮頸癌實(shí)驗(yàn)標(biāo)本及建立宮頸癌臨床病理信息庫;標(biāo)本分組并按要求制作切片(含白片、HE切片及組織芯片制作)。最后應(yīng)用量子點(diǎn)免疫熒光原位雜交技術(shù)(QDs-FISH)檢測(cè)高危型HPV。結(jié)果 QDs-FISH結(jié)合biotin-labeled DNA探針檢測(cè)宮頸(癌)組織,顯示HrHPV陽性信號(hào)位于腫瘤細(xì)胞胞核。在正常宮頸組織中未見Hr-HPV陽性表達(dá);80例宮頸鱗狀細(xì)胞癌細(xì)胞有67例陽性表達(dá),占83.75%;24例腺癌標(biāo)本有18例陽性表達(dá),占66.66%。Hr-HPV僅在癌組織中高表達(dá),與正常宮頸組織相比,差異有統(tǒng)計(jì)學(xué)意義(P0.05);而鱗狀細(xì)胞癌與腺癌組織中的表達(dá)差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 (1)Hr-HPV感染與宮頸癌的發(fā)生發(fā)展密切相關(guān)。(2)QDs-FISH方法取材方便,依從性好,有較高的敏感性和特異性,重復(fù)性好,檢測(cè)設(shè)備也易于操作,可作為篩查宮頸癌的一種較好方法。
[Abstract]:Objective to study the infection status of high-risk HPV in cervical cancer and related tissues, and to evaluate the specificity and sensitivity of the methods used. Methods 124 cases of paraffin-embedded cervical tissues were collected retrospectively. Including 80 cases of cervical squamous cell carcinoma 24 cases of cervical adenocarcinoma and 20 cases of normal cervix tissues or inflammatory cervix tissues resected due to uterine leiomyoma or adenomyosis etc. The cervical cancer specimens were collected and the clinical pathological information bank of cervical cancer was established. The specimens were divided into groups and made into sections (including HE sections and tissue chips) according to the requirements. Finally, high risk HPVs were detected by quantum dot immunofluorescence in situ hybridization (QDs-FISHH). Results QDs-FISH and biotin-labeled DNA probes were used to detect cervical (carcinoma) tissues. There were 67 cases of positive expression of HrHPV in 80 cases of cervical squamous cell carcinoma, 18 cases of positive expression in 24 cases of adenocarcinoma, and 66.66% of Hr-HPV was only overexpressed in cancer tissue, which showed that the positive signal of HrHPV was located in the nucleus of the tumor cell, and there were 67 cases of positive expression of Hr-HPV in 80 cases of cervical squamous cell carcinoma, and 18 cases of positive expression of Hr-HPV in 24 cases of adenocarcinoma. Compared with normal cervical tissues, the difference was statistically significant (P 0.05), but there was no significant difference in the expression of Hr-HPV between squamous cell carcinoma and adenocarcinoma. Conclusion the infection of Hr-HPV is closely related to the occurrence and development of cervical cancer. It has high sensitivity and specificity, good reproducibility and easy to operate, so it can be used as a better method for screening cervical cancer.
