DMT1在人胎盤絨毛膜癌細(xì)胞中表達(dá)的實(shí)驗(yàn)研究

【摘要】:目的進(jìn)一步確定二價(jià)金屬離子轉(zhuǎn)運(yùn)體1(DMT1)在人胎盤絨毛膜癌細(xì)胞(Be Wo)中的表達(dá)定位,以及缺鐵狀態(tài)下m RNA的表達(dá)變化。方法采用免疫細(xì)胞化學(xué)技術(shù)觀察DMT1在Be Wo細(xì)胞中的表達(dá)定位,采用實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)檢測DMT1在Be Wo細(xì)胞缺鐵狀態(tài)下m RNA的表達(dá)變化。結(jié)果 DMT1在Be Wo細(xì)胞中呈陽性表達(dá),其彌漫性表達(dá)定位于細(xì)胞質(zhì)和細(xì)胞膜上;DMT1+IRE m RNA和DMT1-IRE m RNA的相對(duì)表達(dá)量隨缺鐵干預(yù)濃度和時(shí)間的增加而上調(diào)。DMT1+IRE m RNA的相對(duì)表達(dá)量高于DMT1-IRE m RNA。結(jié)論DMT1大量表達(dá)在Be Wo細(xì)胞質(zhì)和細(xì)胞膜上,且表達(dá)受機(jī)體不同鐵水平的調(diào)節(jié),說明其在胎盤鐵轉(zhuǎn)運(yùn)過程中發(fā)揮重要作用,為進(jìn)一步探討胎盤鐵轉(zhuǎn)運(yùn)分子機(jī)制提供實(shí)驗(yàn)基礎(chǔ)。
[Abstract]:Objective to determine the expression localization of divalent metal ion transporter 1 (DMT1) in human placental choriocarcinoma cell line be Woe. Methods Immunocytochemical technique was used to observe the expression localization of DMT1 in be Wo cells. Real-time fluorescence quantitative polymerase chain reaction was used to detect the expression of m RNA in be Wo cells under iron deficiency. Results the expression of DMT1 was positive in be Wo cells. The diffuse expression of DMT1 IRE m RNA and DMT1-IRE m RNA in cytoplasm and cell membrane increased with the increase of iron-deficiency concentration and time. The relative expression of DMT1 IRE m RNA was higher than that of DMT1-IRE m RNA.Conclusion DMT1 is highly expressed in the cell membrane. Be Wo cytoplasm and cell membrane, The expression is regulated by different iron levels, which indicates that it plays an important role in the process of placental iron transport, which provides the experimental basis for further study of the molecular mechanism of placental iron transport.
