【摘要】：目的利用量子點(diǎn)(QDs)免疫熒光技術(shù)對(duì)卵巢癌SKOV3細(xì)胞上葉酸受體(FR)及人附睪蛋白(HE)4進(jìn)行定位,初步揭示QDs多重染色在卵巢癌細(xì)胞體外成像的應(yīng)用價(jià)值。方法采用活潑酯法,構(gòu)建不同發(fā)射波長(zhǎng)的QDs-鏈霉親和素(SA)復(fù)合物,分別與生物素化-葉酸(FA)及生物素化-抗HE4抗體結(jié)合,形成特異性熒光探針,同時(shí)靶向結(jié)合SKOV3細(xì)胞,實(shí)現(xiàn)細(xì)胞雙重染色。結(jié)果構(gòu)建的QD-FA及QD-抗HE4抗體靶向探針具有極強(qiáng)的特異性,量子點(diǎn)多重染色的熒光強(qiáng)度與單重染色相比差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。量子點(diǎn)具有極強(qiáng)的光學(xué)穩(wěn)定性。結(jié)論量子點(diǎn)多重染色在卵巢癌細(xì)胞體外成像中具有一定的應(yīng)用價(jià)值,可為卵巢癌早期檢測(cè)提供一種新思路。
[Abstract]:Objective to localize folic acid receptor (FRR) and human epididymal protein (HE4) on ovarian cancer SKOV3 cells by using quantum dot QDs- immunofluorescence technique, and to reveal the value of QDs multiple staining in imaging ovarian cancer cells in vitro. Methods active ester method was used. QDs- streptavidin (SA) complexes with different emission wavelengths were constructed, which were combined with biotinylated folate (FAA) and biotinylated anti HE4 antibodies to form specific fluorescent probes and target SKOV3 cells. Results the QD-FA and QD- anti HE4 antibody targeting probes were highly specific. There is no significant difference in fluorescence intensity between QDs and QDs compared with single staining. Quantum Dots have strong optical stability. Conclusion Quantum Dot Multiple-staining is valuable in the imaging of ovarian cancer cells in vitro. It may provide a new idea for early detection of ovarian cancer.
