【摘要】：目的：研究IL-24基因聯(lián)合順鉑對宮頸癌裸鼠移植瘤的生長抑制作用以及對腫瘤細(xì)胞侵襲、轉(zhuǎn)移能力的影響。 方法：將構(gòu)建成功的宮頸癌裸鼠模型隨機(jī)分為6組：PBS組（瘤體內(nèi)注射磷酸鹽緩沖液）、pDC316空質(zhì)粒組（瘤體內(nèi)注射脂質(zhì)體包裹的空質(zhì)粒pDC316）、IL-24組（瘤體內(nèi)注射脂質(zhì)體包裹的基因重組質(zhì)粒pDC316-hIL-24）、半量順鉑組（腹腔內(nèi)注射順鉑2.5mg/kg）、全量順鉑組（腹腔內(nèi)注射順鉑5mg/kg）、聯(lián)合組（瘤體內(nèi)注射pDC316-hIL-24聯(lián)合腹腔內(nèi)注射半量順鉑），通過比較各組移植瘤體積和動物體重變化，評價IL-24基因聯(lián)合順鉑的抑瘤效果和藥物副作用，應(yīng)用PCR法測定移植瘤中hIL-24的表達(dá)，westernblot法檢測各組移植瘤組織中β-catenin和E-cadherin的表達(dá)變化，免疫組化方法研究各組淋巴結(jié)轉(zhuǎn)移情況。 結(jié)果：人宮頸癌Siha細(xì)胞裸鼠模型建立成功；PCR結(jié)果表明hIL-24成功轉(zhuǎn)染入腫瘤細(xì)胞并表達(dá)；聯(lián)合組治療后移植瘤體積與全量順鉑組治療后腫瘤體積差別無統(tǒng)計(jì)學(xué)意義（P＞0.05）；聯(lián)合組裸鼠體重明顯大于全量順鉑組（P＜0.05）；聯(lián)合組下調(diào)β-catenin和上調(diào)E-cadherin的表達(dá)效果最明顯，，顯微鏡下觀察見聯(lián)合組淋巴結(jié)中腫瘤細(xì)胞數(shù)較少且有部分壞死。 結(jié)論：IL-24基因聯(lián)合半量順鉑治療能達(dá)到全量順鉑的抑瘤效果，減少化療藥物的使用劑量；IL-24基因可通過下調(diào)β-catenin和上調(diào)E-cadherin的表達(dá)水平進(jìn)而抑制Siha腫瘤細(xì)胞侵襲和轉(zhuǎn)移；聯(lián)合治療比單獨(dú)使用IL-24或順鉑能更大程度抑制Siha細(xì)胞侵襲和轉(zhuǎn)移。
[Abstract]:Aim: to study the effects of IL-24 gene combined with cisplatin on the growth inhibition and invasion and metastasis of cervical carcinoma xenografts in nude mice. Methods: a successful nude mouse model of cervical cancer was randomly divided into 6 groups: 1: PBS group (pDC316 empty plasmid group was injected with phosphate buffer solution in the tumor (pDC316) empty plasmid pDC316- IL-24 was injected into the tumor body (liposome encapsulated group was injected into the tumor body). The recombinant plasmid pDC316-hIL-24, half dose cisplatin group (intraperitoneal injection of cisplatin 2.5 mg / kg), whole dose group (intraperitoneal injection of cisplatin 5 mg / kg), combined group (intratumoral injection of pDC316-hIL-24 and intraperitoneal injection of half dose of cisplatin) were used to compare the volume of transplanted tumor and the weight of animals. To evaluate the anti-tumor effect and side effect of IL-24 combined with cisplatin, the expression of hIL-24 in transplanted tumor was detected by PCR method and the expression of 尾 -catenin and E-cadherin was detected by Western blot. The lymph node metastasis in each group was studied by immunohistochemical method. Results: human cervical cancer Siha cells were successfully transfected into tumor cells and expressed in nude mice. There was no significant difference in tumor volume between the combined group and the whole cisplatin group (P > 0.05), the weight of nude mice in the combined group was significantly higher than that in the whole cisplatin group (P < 0.05), the effect of down-regulating 尾 -catenin and up-regulation of E-cadherin was the most obvious in the combined group. It was observed under microscope that the number of tumor cells in the combined group was small and there was partial necrosis. Conclusion the tumor inhibition effect of total cisplatin can be achieved by the combination of 1: IL-24 gene and half dose cisplatin. Reducing the dosage of chemotherapeutic drugs can inhibit the invasion and metastasis of Siha tumor cells by down-regulating the expression of 尾 -catenin and up-regulating the expression of E-cadherin. Combination therapy can significantly inhibit the invasion and metastasis of Siha cells compared with IL-24 or cisplatin alone.
