【摘要】:目的 探討甲基化轉(zhuǎn)移酶抑制劑BIX01294(BIX)對小鼠子宮內(nèi)膜基質(zhì)細(xì)胞遷移和蛻膜化過程的作用和影響。方法 取妊娠第3.5天小鼠子宮分離培養(yǎng)得小鼠子宮內(nèi)膜基質(zhì)細(xì)胞,采用細(xì)胞劃痕運動分析、體外誘導(dǎo)小鼠子宮內(nèi)膜基質(zhì)細(xì)胞蛻膜化模型及Real-time PCR方法分別從形態(tài)學(xué)和分子生物學(xué)角度觀察BIX對小鼠子宮內(nèi)膜基質(zhì)細(xì)胞遷移及蛻膜化的影響。結(jié)果 在一定濃度范圍內(nèi)(BIX≤15μmol/L),小鼠子宮內(nèi)膜基質(zhì)細(xì)胞的遷移距離隨著BIX濃度的升高而增加,與對照組相比,BIX 15μmol/L處理組基質(zhì)細(xì)胞的遷移距離增加最明顯,差異具有統(tǒng)計學(xué)意義,當(dāng)BIX濃度提高到20μmol/L時,細(xì)胞遷移距離增加不明顯,且此時開始出現(xiàn)細(xì)胞的死亡;與對照組相比,BIX處理24 h組小鼠子宮內(nèi)膜基質(zhì)細(xì)胞Ehmt2 m RNA表達(dá)顯著下調(diào)(P0.01);并且15μmol/L BIX能夠抑制小鼠子宮內(nèi)膜基質(zhì)細(xì)胞蛻膜化反應(yīng)。結(jié)論 一定濃度范圍的BIX通過抑制Ehmt2m RNA在基質(zhì)細(xì)胞中的表達(dá)促進(jìn)小鼠子宮內(nèi)膜基質(zhì)細(xì)胞的遷移,并且對小鼠子宮基質(zhì)細(xì)胞的蛻膜化過程有抑制作用。
[Abstract]:Objective to investigate the effects of BIX01294 (BIX01294) on the migration and decidualization of mouse endometrial stromal cells. Methods Mouse endometrial stromal cells were isolated and cultured on day 3.5 of pregnancy. Cell scratch motion analysis was used. The model of decidualization induced by mouse endometrial stromal cells in vitro and Real-time PCR method were used to observe the effects of BIX on migration and decidualization of mouse endometrial stromal cells from the point of view of morphology and molecular biology.
