【摘要】：目的:探討納米雄黃對不同宮頸癌細胞株Hela(HPV18陽性,腺癌),Caski(HPV16陽性,鱗癌),C33A(HP V陰性,腺癌)增殖、凋亡的影響。方法:體外培養(yǎng)3種不同宮頸癌細胞,采用不同質(zhì)量濃度(5,10,2 0,4 0 mg/L)納米雄黃作用于不同宮頸癌細胞2 4,4 8,7 2,9 6 h,相差顯微鏡下觀察不同質(zhì)量濃度組與對照組細胞形態(tài)學(xué)變化;MTT法測定藥物對細胞生長的抑制作用;流式細胞儀檢測細胞凋亡及細胞周期。結(jié)果:納米雄黃混懸液(5,10,20,40 mg/L)作用于Caski,Hela,C33A細胞24,48,72,96 h,均可抑制細胞增殖,且呈時間劑量依賴性,抑制率波動于9.02%~49.06%,以20 mg/L質(zhì)量濃度組為例,抑制率Caski(39.15%)Hela(36.17%)C33A(30.56%)。隨著質(zhì)量濃度的增加,早期凋亡、中晚期凋亡細胞均增加,凋亡率(早期+中晚期凋亡率)波動于19.29%~99.54%,以20 mg/L質(zhì)量濃度組為例,凋亡率Caski(60.43±2.88)%Hela(41.95±3.01)%C33A(43.49±2.19)%。細胞周期結(jié)果顯示高質(zhì)量濃度組(20,40 mg/L)對Hela,Caski細胞有G2/M期阻滯。結(jié)論:納米雄黃混懸液可抑制不同宮頸癌細胞株的增殖,促進細胞凋亡,其對Caski細胞的作用最強,對Hela細胞次之,對C33A細胞相對最弱。對HPV陽性的細胞可產(chǎn)生G2/M期阻滯。
[Abstract]:Objective: to investigate the effects of nano-realgar on the proliferation and apoptosis of different cervical cancer cell lines (Hela(HPV18 positive, Casikius HPV16 positive, squamous cell carcinoma C33AHPV negative, adenocarcinoma). Methods: three kinds of cervical cancer cells were cultured in vitro. Nano realgar was used to treat different cervical cancer cells with different concentration of 1010 mg / L (40 mg / L) for 6 h. The cell morphological changes of different concentration groups and control group were observed under phase contrast microscope and MTT assay was used to determine the inhibitory effect of the drug on cell growth. Cell apoptosis and cell cycle were measured by flow cytometry. Results: the cell proliferation was inhibited in a time and dose-dependent manner when treated with nano-realgar suspension (51010C / L) for 2448C 33A cells for 24 h, and the inhibition rate fluctuated from 9.02 to 49.065.In the 20 mg/L mass concentration group, the cell proliferation was inhibited in a dose-dependent manner, and the inhibition rate varied from 9.02to 49.06mg / L, especially in the 20 mg/L group, and the proliferation of the cells was inhibited in a dose-dependent manner. With the increase of mass concentration, early apoptosis and late apoptotic cells increased, and the apoptotic rate (early, late stage apoptosis rate) fluctuated at 19.29 ~ 99.54%, taking the 20 mg/L mass concentration group as an example. The cell cycle results showed that the high concentration group (20 mg / L) had G 2 / M phase arrest on Hela Caski cells. Conclusion: nano realgar suspension can inhibit the proliferation of different cervical cancer cell lines and promote apoptosis, and its effect on Caski cells is the strongest. For Hela cells, it was the weakest for C33A cells, and G2 / M phase arrest was found for HPV positive cells.
