【摘要】：目的觀察人肌母細(xì)胞瘤體注射治療人宮頸癌細(xì)胞裸鼠移植瘤的效果,并與順鉑進(jìn)行比較。方法裸鼠45只,皮下注射He La細(xì)胞制作宮頸癌細(xì)胞移植瘤,隨機(jī)分為人肌母細(xì)胞組、順鉑組、生理鹽水組各15只,分別采取人肌母細(xì)胞瘤體注射、順鉑腹腔注射、生理鹽水瘤體注射,用藥后連續(xù)觀察4周。于用藥前后觀察裸鼠一般情況,測(cè)算裸鼠體質(zhì)量及腫瘤體積;采用HE染色觀察各組腫瘤組織形態(tài)及細(xì)胞形態(tài);分別采用免疫組化法、Western blotting法檢測(cè)腫瘤組織中的p53蛋白。結(jié)果人肌母細(xì)胞組裸鼠用藥后體質(zhì)量較治療前增高,腫瘤體積較用藥前縮小,且用藥后腫瘤體積小于其余兩組(P均0.05)。人肌母細(xì)胞組裸鼠用藥后的反應(yīng)性、精神、飲食狀況均優(yōu)于其余兩組。病理檢查示生理鹽水組細(xì)胞異型性明顯、腫瘤細(xì)胞體積大、細(xì)胞核增多;人肌母細(xì)胞組可見(jiàn)大量凋亡壞死的腫瘤細(xì)胞,大量細(xì)胞固縮、細(xì)胞核碎裂,腫瘤組織纖維化明顯;順鉑組腫瘤細(xì)胞固縮、細(xì)胞核裂解不明顯,腫瘤組織纖維化程度不顯著。人肌母細(xì)胞組、順鉑組、生理鹽水組腫瘤組織中p53蛋白陽(yáng)性表達(dá)率分別為20.0%、33.3%、60.0%,p53蛋白相對(duì)表達(dá)量分別為0.321±0.054、0.379±0.067、0.534±0.064,三組相比,P均0.05。結(jié)論人肌母細(xì)胞瘤體注射可減小人宮頸癌細(xì)胞裸鼠移植瘤的體積,促使腫瘤細(xì)胞凋亡(機(jī)制可能與下調(diào)p53蛋白表達(dá)有關(guān)),抑瘤效果優(yōu)于順鉑。
[Abstract]:Objective to observe the effect of human myoblast tumor injection on human cervical carcinoma xenografts in nude mice, and compare it with cisplatin. Methods 45 nude mice were injected subcutaneously with he La cells to make cervical carcinoma transplanted tumor, and they were randomly divided into human myoblast group. Cisplatin group (n = 15) and saline group (n = 15) were injected with human myoblast, intraperitoneal injection of cisplatin and injection of normal saline respectively. The body mass and tumor volume of nude mice were measured and the morphology of tumor tissues and cells in each group were observed by HE staining. P53 protein in tumor tissue was detected by immunohistochemistry and Western blotting. Results the body mass of nude mice in human myoblast group was higher than that before treatment, and the tumor volume was smaller than that before treatment. The tumor volume was smaller than that of the other two groups (P 0.05). The reactivity, spirit and diet of nude mice in the human myoblast group were better than those in the other two groups. The pathological examination showed that the cells in the saline group were obviously abnormal and the tumor cells were large. Large numbers of apoptotic and necrotic tumor cells were observed in the human myoblast group, with large numbers of cells pyknosis, nuclear fragmentation, and tumor tissue fibrosis, while in the cisplatin group, the tumor cells were pyknotic, and the nucleus cleavage was not obvious. The degree of fibrosis in tumor tissue was not significant. Human myoblast group, cisplatin group, The positive expression rates of p53 protein in normal saline group were 20.0 / 33.3and 0.321 鹵0.054 + 0.379 鹵0.067 鹵0.534 鹵0.064, respectively. Conclusion Human myoblast tumor injection can reduce the volume of human cervical carcinoma xenografts in nude mice. The mechanism may be related to the down-regulation of p53 protein expression, and the inhibitory effect is better than cisplatin.
