IL-11Rα介導(dǎo)的分子探針在宮頸癌模型中的顯像研究

【摘要】:目的:探討IL~(-1)1Rα介導(dǎo)的核素標(biāo)記探針~(99)Tc~m-DTPA-c(CGRRAGGSC)在宮頸癌模型中的顯像特性。方法:采用Western Blot法檢測(cè)宮頸癌Hela細(xì)胞株IL~(-1)1Rα表達(dá)水平,免疫熒光進(jìn)行HELA細(xì)胞體外結(jié)合實(shí)驗(yàn);然后合成核素標(biāo)記探針,對(duì)其標(biāo)記率及穩(wěn)定性進(jìn)行檢測(cè),最后進(jìn)行荷瘤裸鼠模型SPECT顯像。結(jié)果:IL~(-1)1Rα在HELA細(xì)胞中呈明顯高表達(dá);合成探針即刻標(biāo)記率達(dá)96.62%,放置于室溫和人新鮮血清孵育12 h后放射化學(xué)純度仍在90%;通過(guò)瘤體間質(zhì)注射~(99)Tc~m-DTPA-c(CGRRAGGSC)見(jiàn)腫瘤局部呈明顯的放射性濃聚且持續(xù)較長(zhǎng)時(shí)間,而游離~(99)Tc~mO_4~-注射于瘤體內(nèi)30 min即見(jiàn)腫瘤濃聚部位淺淡。結(jié)論:標(biāo)記探針可由IL~(-1)1Rα介導(dǎo)對(duì)宮頸癌進(jìn)行特異靶向性顯像,有望為高表達(dá)IL~(-1)1Rα的惡性腫瘤提供早期、特異診斷及靶向治療。
[Abstract]:Objective: to investigate the IL~(-1)1R 偽 -mediated radionuclide labeled probe TcmDTPA-cr rRAGSCC. Methods: Western Blot assay was used to detect the expression of IL~(-1)1R 偽 in cervical cancer Hela cell line. Immunofluorescence assay was performed on HELA cells in vitro. Then the radionuclide labeled probe was synthesized and its labeling rate and stability were tested. Finally, SPECT imaging of the nude mice bearing tumor was performed. Results the expression of 1 R 偽 in HELA cells was significantly higher than that in the control group. The immediate labeling rate of the synthetic probe was 96.62%, and the radiochemical purity was still 90 after being incubated at room temperature with human fresh serum for 12 hours. Through interstitial injection of TcmDTPA-cGSCC, it was found that the tumor area was radioactive and concentrated and lasted for a long time. However, 30 min after injection into the tumor body, the concentration of the tumor was observed. Conclusion: the labeled probe can be labeled by ILO-1). 1R 偽 mediated specific targeting imaging of cervical cancer. It is expected to provide early, specific diagnosis and targeted therapy for malignant tumors with high expression of IL~(-1)1R 偽.
