【摘要】：目的檢測miR-20a和let-7a在卵巢癌患者血漿中的表達(dá),探討其臨床意義。方法選取卵巢癌患者50例為實(shí)驗(yàn)組,另選擇同期與其年齡性別相匹配的健康體檢者20例為健康對(duì)照組,采用實(shí)時(shí)定量熒光定量酶鏈反應(yīng)(qRT-PCR)檢測血漿miR-20a和let-7a表達(dá)水平,同時(shí)采用電化學(xué)發(fā)光法測定其血漿中糖類抗原125(CA125)的含量并進(jìn)行統(tǒng)計(jì)分析。結(jié)果卵巢癌患者血漿中miR-20a和let-7a的相對(duì)表達(dá)量分別為(4.02±3.77)、(0.42±0.34),卵巢癌患者血漿中miR-20a和CA125的表達(dá)均高于對(duì)照組(P0.01),let-7a表達(dá)低于對(duì)照組(P0.01)。臨床病理分期Ⅰ~Ⅱ期和Ⅲ~Ⅳ期卵巢癌患者血漿let-7a相對(duì)表達(dá)量的中位數(shù)分別為0.39、0.21,差異有統(tǒng)計(jì)學(xué)意義(P0.01),而血漿miR-20a的表達(dá)與臨床病理分期無關(guān)(P0.05),miR-20a和let-7a的相對(duì)表達(dá)量與年齡、淋巴結(jié)轉(zhuǎn)移和組織學(xué)分類均無關(guān)(P0.05)。miR-20a、let-7a和CA125的ROC曲線下面積為0.898(95%CI:0.802~0.957)、0.876(95%CI:0.775~0.942)、0.819(95%CI:0.708~0.900)。結(jié)論卵巢癌患者血漿中miR-20a和let-7a均表達(dá)異常,提示能夠成為輔助診斷卵巢癌的標(biāo)志物。
[Abstract]:Objective to detect the expression of miR-20a and let-7a in plasma of patients with ovarian cancer and to explore its clinical significance. Methods 50 patients with ovarian cancer were selected as experimental group and 20 healthy persons matched with their age and sex as healthy control group. The expression of miR-20a and let-7a in plasma was detected by real-time quantitative fluorescence quantitative polymerase chain reaction (qRT-PCR). The relative expression of miR-20a and let-7a in plasma of patients with ovarian cancer was 4.02 鹵3.77 鹵0.42 鹵0.34, respectively. The expression of miR-20a and CA125 in plasma of patients with ovarian cancer was both determined by electrochemiluminescence.
