兩種冷凍方法對牛卵巢組織竇前卵泡活性的影響

【摘要】:目的:探討兩種冷凍方法對牛卵巢組織竇前卵泡活性的影響。方法:取新鮮的牛卵巢組織隨機分為新鮮對照組、程序化冷凍組和玻璃化冷凍組,分別加入Liberase DH酶消化,然后挑取竇前卵泡并分類計數,用calcein AM以及ethidium homodimer對竇前卵泡進行熒光染色,鑒定竇前卵泡的活性。結果:3組各級卵泡的數量及分布差異無統(tǒng)計學意義(P=0.765);與新鮮對照組(91%)相比,程序化冷凍組和玻璃化冷凍組竇前卵泡中未受損卵泡所占比例(80%和78%)略有降低(P=0.012)。3組中所占比例占優(yōu)勢的始基卵泡中未受損卵泡率差異無統(tǒng)計學意義(P=0.079);與新鮮對照組相比,兩冷凍組初級卵泡中未受損卵泡率有所降低(P0.05),但兩冷凍組間差異無統(tǒng)計學意義(P0.05)。結論:程序化冷凍法和玻璃化冷凍法均能較好地保存卵巢組織竇前卵泡的活性。
[Abstract]:Objective: to investigate the effect of two freezing methods on the activity of bovine ovarian preantral follicles. Methods: fresh bovine ovarian tissues were randomly divided into three groups: fresh control group, programmed freezing group and vitrification group. After digesting with Liberase DH enzyme, preantral follicles were collected and counted. The preantral follicles were stained with calcein AM and ethidium homodimer. Results there was no significant difference in the number and distribution of follicles in the three groups (P < 0. 765). Compared with fresh control group (91%). The percentage of undamaged follicles in preantral follicles in programmed and vitrified groups was 80% and 78%, respectively. There was no significant difference in the percentage of undamaged follicles in primordial follicles in group 3 (P 0.079). Compared with the fresh control group, the rate of undamaged follicles in the primary follicles in the two freezing groups was lower than that in the fresh control group (P 0.05). But there was no significant difference between the two groups. Conclusion: programmed freezing and vitrification can better preserve the activity of preantral follicles in ovarian tissue.
