【摘要】：目的探討miRNA成熟關(guān)鍵基因DICER、DROSHA和RAN的基因多態(tài)性與女性復(fù)發(fā)流產(chǎn)的相關(guān)性。方法收集2013年6月至2015年12月于四川大學(xué)華西第二醫(yī)院就診的不明原因復(fù)發(fā)流產(chǎn)(URSA)患者共217例,至少有1次生育史的育齡婦女為正常對照組390例,對照組為同期進(jìn)行體檢和孕前檢查育齡婦女。采用病例對照研究的方法,應(yīng)用PCR-限制性片段長度多態(tài)性技術(shù)檢測miRNA成熟過程中關(guān)鍵基因DICER rs3742330、DROSHArs10719和RAN rs14035單核苷酸多態(tài)性。結(jié)果 DICERrs3742330,DROSHArs10719和RANrs14035各等位基因和基因型頻率在URSA組和對照組差異無統(tǒng)計學(xué)意義(P0.05)。聯(lián)合分析發(fā)現(xiàn),DICERrs3742330/DROSHArs10719GG/TT+TC協(xié)同會增加復(fù)發(fā)流產(chǎn)的風(fēng)險(OR=1.657,95%CI=1.006~2.731,P=0.047)。雖然DICERrs3742330/RANrs14035GG/TT+TC在URSA組較對照組增高,但差異無統(tǒng)計學(xué)意義(OR=1.977,95%CI=0.956~4.087,P=0.066),而DROSHA10719/RAN14035TT+TC/TT+TC與復(fù)發(fā)流產(chǎn)無相關(guān)性(OR=0.958,95%CI=0.679~1.353,P=0.808)。結(jié)論DICERrs3742330聯(lián)合DROSHA rs10719可能與URSA相關(guān)。
[Abstract]:Objective to investigate the key gene DICER of miRNA maturation. DROSHA and RAN gene polymorphisms were associated with recurrent abortion in women. Methods from June 2013 to December 2015, unknown causes of recurrent abortion were collected from Huaxi second Hospital, Sichuan University. (. There were 217 patients with URSA. 390 women of childbearing age with at least one history of childbearing were in the control group, and those in the control group underwent physical examination and pre-pregnancy examination at the same time. A case-control study was used. PCR- restriction fragment length polymorphism (PCR) was used to detect the key gene DICER rs3742330 during miRNA maturation. DROSHArs10719 and RAN rs14035 single nucleotide polymorphisms. Results DICERrs3742330. The frequencies of alleles and genotypes of DROSHArs10719 and RANrs14035 were not significantly different between URSA group and control group (P 0.05). DICERrs3742330/DROSHArs10719GG/TT TC synergism increased the risk of recurrent abortion. [95] CII 1.006 / 2.731. Although DICERrs3742330/RANrs14035GG/TT TC in URSA group was higher than that in control group. However, the difference was not statistically significant (1.977 / 95 CI = 0.956 / 4.087 / P0.066). There was no correlation between DROSHA10719/RAN14035TT TC/TT TC and recurrent abortion. Conclusion DICERrs3742330 combined with DROSHA rs10719 may correlate with URSA.
