【摘要】:目的:尋找上皮性卵巢癌(epithelial ovarian cancer,EOC)癌相關(guān)成纖維細(xì)胞(cancer associated fibroblasts,CAFs)和正常成纖維細(xì)胞(normal fibroblasts,NFs)中差異表達(dá)的m RNA并進(jìn)行分析,為進(jìn)一步闡明腫瘤間質(zhì)中癌相關(guān)成纖維細(xì)胞影響卵巢癌細(xì)胞惡性行為的機(jī)制提供依據(jù)。方法:分別提取3例上皮性卵巢癌的CAFs及3例正常卵巢組織的NFs,抽提總RNA,進(jìn)行全基因表達(dá)譜分析。對差異基因進(jìn)行基因本體論(Gene Ontology,GO)富集及信號通路聯(lián)合分析,推定相關(guān)差異基因生物學(xué)功能,并對MDM2、NR3C1等6個上調(diào)的差異基因進(jìn)行熒光定量PCR驗證。結(jié)果:對芯片結(jié)果的分析顯示差異基因共1 636個。q RTPCR對MDM2、NR3C1等6個差異基因的驗證結(jié)果與芯片結(jié)果相符,生物信息學(xué)分析顯示上述差異基因與癌相關(guān)通路、肌動蛋白細(xì)胞骨架重塑通路等有關(guān),提示可能參與了對卵巢癌細(xì)胞惡性行為的促進(jìn)作用。結(jié)論:EOC腫瘤間質(zhì)中的成纖維細(xì)胞存在調(diào)控癌細(xì)胞的差異基因,分析這些差異基因為進(jìn)一步研究卵巢癌成纖維細(xì)胞調(diào)控癌細(xì)胞生物學(xué)行為的分子機(jī)制提供了基本依據(jù)。
[Abstract]:Objective: to search for epithelial ovarian cancer (epithelial ovarian, cancer, EOC) cancer associated fibroblasts (cancer associated, fibroblasts, CAFs) and normal fibroblasts (normal, fibroblasts, NFs) in the differential expression of M RNA and analysis, provides the basis for further clarify the tumor stroma in cancer related to the malignant behavior of ovarian cancer cells the mechanism of the influence of fiber cells. Methods: CAFs were extracted from 3 cases of epithelial ovarian cancer and 3 cases of normal ovarian tissue NFs, extract the total RNA spectrum to analyze the gene expression of different genes. Gene Ontology (Gene Ontology, GO) and signaling pathway enrichment analysis, correlation between the putative gene biological functions and, MDM2, fluorescence quantitative PCR validation of differential gene NR3C1 6 up-regulated. Results: analysis of the microarray results showed differences in 1636 genes of.Q RTPCR MDM2, gene NR3C1 6 differences The results are consistent with the microarray results, bioinformatics analysis showed that the difference of gene and cancer related pathways, actin cytoskeleton remodeling pathway, suggesting that it may be involved in the malignant behavior of ovarian cancer cells. Conclusion: EOC tumor interstitial fibroblast gene is differentially regulated in cancer cells, analysis the basis for these differential genes for further research of the molecular mechanism of ovarian cancer cells into fibroblasts regulates the biological behavior of cancer.
