【摘要】：目的:通過檢測不同胚胎質(zhì)量的體外受精(in vitro fertilization,IVF)患者精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375的表達(dá)情況,探討精子來源的mi RNAs與胚胎質(zhì)量的相關(guān)性。方法:選擇2012年9月至12月在中南大學(xué)湘雅醫(yī)院生殖醫(yī)學(xué)中心行IVF治療原發(fā)不育的男性患者44例,收集IVF取卵當(dāng)日剩余新鮮精液標(biāo)本,采用實(shí)時(shí)熒光定量PCR測定精子mi RNAs(mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375)的表達(dá)水平。觀察胚胎情況后分組,第3天胚胎評分的平均值8分為實(shí)驗(yàn)組,≥8分為對照組。比較實(shí)驗(yàn)組和對照組患者一般情況及實(shí)驗(yàn)室資料,分析精子mi RNAs表達(dá)水平與胚胎質(zhì)量的相關(guān)性。結(jié)果:實(shí)驗(yàn)組精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375表達(dá)水平低于對照組(P0.01)。實(shí)驗(yàn)組與對照組的獲卵數(shù)、減數(shù)分裂II期(metaphase II,MII)卵子數(shù)、雙原核(dual pronuclear,2PN)受精數(shù)、卵裂數(shù)、受精率差異無統(tǒng)計(jì)學(xué)意義(P0.05);實(shí)驗(yàn)組卵裂率低于對照組(P0.05)。精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375表達(dá)水平與第2,3天胚胎碎片率呈負(fù)相關(guān),與第3天胚胎卵裂球數(shù)呈正相關(guān),與胚胎評分呈正相關(guān)。結(jié)論:精子mi RNA-21,mi RNA-34c,mi RNA-140,mi RNA-375的表達(dá)水平上升可能影響卵裂期胚胎質(zhì)量,對胚胎發(fā)育可能起一定的積極作用。
