【摘要】:目的:探討4.1N蛋白對卵巢透明細胞癌細胞遷移和侵襲能力的調(diào)控并初步探討相關分子機制。方法:應用免疫組化法檢測4.1N蛋白在卵巢透明細胞癌組織中的表達缺失狀況。建立4.1N穩(wěn)定轉(zhuǎn)染卵巢透明細胞癌ES-2細胞系,通過劃痕實驗、Transwell遷移和侵襲實驗觀察細胞遷移侵襲能力的改變。利用qRT-PCR和Western blot法檢測緊密連接相關分子(Claudin-4和ZO-1)、上皮-間質(zhì)轉(zhuǎn)化(EMT)標記物(E-cadherin、Ncadherin和Vimentin)和相關轉(zhuǎn)錄因子(Twist、Slug和ZEB-2)及基質(zhì)金屬蛋白酶(MMP-2和MMP-3)表達水平的變化。結(jié)果:與卵巢異位的子宮內(nèi)膜組織相比,卵巢透明細胞癌中4.1N蛋白表達水平顯著降低甚至缺失(P0.0001)。過表達4.1N后,ES-2細胞的遷移和侵襲能力顯著降低,Claudin-4和ZO-1表達上調(diào),Twist、Slug、ZEB-2、MMP-2和MMP-3表達水平下降。結(jié)論:4.1N在卵巢透明細胞癌細胞中可能通過調(diào)控緊密連接蛋白、部分上皮-間質(zhì)轉(zhuǎn)化轉(zhuǎn)錄因子和基質(zhì)金屬蛋白酶的表達水平,抑制癌細胞的遷移和侵襲能力,進而參與負性調(diào)控卵巢透明細胞癌的演進。
[Abstract]:Objective: to investigate the regulation of 4.1 N protein on the migration and invasion of ovarian clear cell carcinoma cells and to explore the molecular mechanism. The expression of 4.1N protein in clear cell carcinoma of ovary was detected by immunohistochemical method. The ES-2 cell line of ovarian clear cell carcinoma was established by 4.1N stable transfection. Through the scratch test. Transwell migration and invasion assay were used to observe the change of cell migration and invasion ability. QRT-PCR and Western blot were used to detect the tightly linked molecules (. Claudin-4 and ZO-1). E-cadherin and Vimentin (E-cadherin and Vimentin) and related transcription factor Twist. Slug and ZEB-2) and matrix metalloproteinase (MMP-2) and MMP-3) expression levels. Results: compared with ovarian ectopic endometrium tissue. The expression level of 4.1N protein in ovarian clear cell carcinoma was significantly decreased or even deleted P0.0001N, and the migration and invasion ability of Ovarian clear cell carcinoma was significantly decreased after the expression of 4.1 N. The expression of Claudin-4 and ZO-1 up-regulated the expression of Twistus Slugfus ZEB-2. Conclusion the expression of MMP-2 and MMP-3 in ovarian clear cell carcinoma cells may be regulated by tight junction protein. The expression of transcriptional factors and matrix metalloproteinases (MMPs) in some epithelium-mesenchymal cells inhibits the migration and invasion of cancer cells and thus participates in the negative regulation of the progression of ovarian clear cell carcinoma.
