維生素D調(diào)節(jié)血管內(nèi)皮生長因子在支氣管肺發(fā)育不良新生大鼠中的作用及機制研究

目的:研究維生素D(VD)及其受體(VDR)通過調(diào)節(jié)VEGF對新生大鼠支氣管肺發(fā)育不良(BPD)的調(diào)節(jié)作用和機制。方法:構(gòu)建BDP模型新生大鼠,分為對照組和BDP組。培養(yǎng)大鼠肺上皮Ⅱ型細胞(RLE-6TN),RLE-6TN細胞分為VDR過表達(Le-VDR慢病毒感染)、VDR過表達對照組(Le-Ctrl)、VDR敲低(Sh-VDR感染)、VDR敲低對照組(Sh-Ctrl),VD單獨處理,VEGF-A沉默組(si VEGF-A); Le-VDR與si VEGF-A共處理(Le-VDR+si VEGF-A);通過CCK-8檢測、流式細胞術(shù)、傷口愈合檢測和體外粘附檢測明確VD和VDR對肺泡上皮細胞的調(diào)節(jié)作用。RTq PCR檢測VDR和VEGF-A mRNA水平,Western blot檢測VDR和VEGF-A的蛋白水平。結(jié)果:與對照相比,BPD新生大鼠的VD水平和VDR表達降低(P<0.01);在RLE-6TN細胞中過表達VDR,細胞增殖、遷移、粘附能力增加(均P<0.01),凋亡率減少(P <0.01);細胞中敲低VDR效果相反,VD刺激后細胞凋亡被抑制,細胞增殖、遷移和...
