DES及INSL3對(duì)體外培養(yǎng)小鼠睪丸引帶細(xì)胞中LGR8轉(zhuǎn)錄和表達(dá)的影響

目的:近年來研究表明環(huán)境雌激素(EEs)對(duì)男性生殖系統(tǒng)的影響日益明顯,且大多數(shù)與睪丸的下降不全及發(fā)育不良有關(guān),而睪丸引帶在睪丸的下降和發(fā)育過程中一直被認(rèn)為扮演著重要角色。研究證實(shí)胰島素樣因子3(INSL3)參與引帶發(fā)育、睪丸下降的過程。INSL3通過與其特異性受體LGR8結(jié)合而在第一階段(經(jīng)腹下降期)發(fā)揮主要作用,且LGR8高表達(dá)于睪丸引帶之上,但有關(guān)其具體作用機(jī)制尚未完全研究清楚。本實(shí)驗(yàn)通過體外培養(yǎng)小鼠睪丸引帶細(xì)胞,利用不同濃度己烯雌酚(DES)和INSL3直接作用于培養(yǎng)的小鼠睪丸引帶細(xì)胞,以探討DES和INSL3對(duì)小鼠睪丸引帶細(xì)胞中LGR8 mRNA及蛋白表達(dá)的影響及其與小鼠睪丸下降的可能關(guān)系。材料與方法:將剛出生3日齡的雄性昆明小鼠顯微剖出其睪丸引帶組織并進(jìn)行細(xì)胞培養(yǎng)。傳代培養(yǎng)后隨機(jī)分組,按DES濃度分為10μg/ml、1μg/ml、0.1μg/ml、0.01μg/ml、DMSO溶劑對(duì)照組和正常對(duì)照組;按INSL3濃度分為20ng/ml、2ng/ml、0.2ng/ml、0.02ng/ml和正常對(duì)照組。在倒置顯微鏡下觀察細(xì)胞形態(tài)并應(yīng)用免疫熒光對(duì)LGR8進(jìn)行亞細(xì)胞定位。加藥持續(xù)作用4...
