小膠質(zhì)細(xì)胞活化在鉛誘導(dǎo)學(xué)習(xí)記憶損傷中的作用及機制探討

【背景】鉛是嚴(yán)重危害我國兒童健康的主要環(huán)境化學(xué)危險因素,具有流行范圍廣、毒性隱匿持久、主要損傷兒童智力發(fā)育等特點。我國目前約有10%~15%的城市兒童血鉛超標(biāo),部分工礦企業(yè)周邊地區(qū)更為嚴(yán)重,甚至發(fā)生多起嚴(yán)重的鉛污染和兒童鉛中毒事件,嚴(yán)重影響了社會安定和兒童身心健康。因此,揭示鉛誘導(dǎo)海馬神經(jīng)元損傷以及學(xué)習(xí)記憶能力下降的細(xì)胞和分子機制,提出有效的防治措施,是目前急需解決的問題。小膠質(zhì)細(xì)胞是中樞神經(jīng)系統(tǒng)(central nervous system,CNS)的一個重要的免疫駐留細(xì)胞。小膠質(zhì)細(xì)胞對外界環(huán)境刺激非常敏感,在許多微環(huán)境因子發(fā)生變化時,小膠質(zhì)細(xì)胞可以迅速做出反應(yīng),表現(xiàn)為從帶有分支的靜息狀態(tài)轉(zhuǎn)變?yōu)榫哂凶冃魏屯淌勺饔玫募せ顮顟B(tài),并可以顯著上調(diào)許多免疫炎性因子的分泌水平,如腫瘤壞死因子α(tumor necrosis factor-α, TNF-α)、白介素-lβ(interleukin-1β, IL-1β)等等。激活的小膠質(zhì)細(xì)胞還能上調(diào)一些炎性相關(guān)因子如iNOS的表達(dá),這些因素被認(rèn)為在介導(dǎo)神經(jīng)元的損害中起著重要的作用。有研究發(fā)現(xiàn)通過脂多糖刺激小膠質(zhì)細(xì)胞活化可以誘導(dǎo)學(xué)習(xí)記憶能力下降,而我們...
