促紅細(xì)胞生成素減輕新生兒窒息后血清誘導(dǎo)HK-2細(xì)胞凋亡作用的胞內(nèi)信號(hào)傳導(dǎo)機(jī)制研究

目的:研究促紅細(xì)胞生成素減輕新生兒窒息后血清誘導(dǎo)HK-2細(xì)胞凋亡作用的胞內(nèi)信號(hào)傳導(dǎo)機(jī)制。方法:(1)以HK-2細(xì)胞(Human renal proximal tubular cell,人近曲腎小管上皮細(xì)胞)為研究對(duì)象。(2)以新生兒重度窒息(Apgar評(píng)分小于4分)后第一天血清(20%體積分?jǐn)?shù))作為攻擊因素。(3)實(shí)驗(yàn)分四組,即對(duì)照組、窒息組、EPO(erythropoietin,促紅細(xì)胞生成素)干預(yù)組、EPO+5-HD (5-Hydroxydecanoic acid sodium salt,5-羥葵酸鹽)干預(yù)組。(4)檢測(cè)指標(biāo):倒置相差顯微鏡下觀察HK-2細(xì)胞生長情況;免疫組織化學(xué)方法檢測(cè)HK-2細(xì)胞胞漿內(nèi)Omi/HtrA2、Caspase-3、XIAP的表達(dá)情況;流式細(xì)胞儀檢測(cè)細(xì)胞凋亡率;激光共聚焦顯微鏡檢測(cè)HK-2細(xì)胞線粒體膜電位(△ψm )變化以及觀察間接免疫熒光雙染色Omi/HtrA2在細(xì)胞內(nèi)從線粒體向胞漿的轉(zhuǎn)位。(5)統(tǒng)計(jì)學(xué)分析:數(shù)據(jù)均用x±s表示,采用one-way ANOVA分析,組間比較用LSD法。用SPSS13.0軟件進(jìn)行處理。P <0.05為差異具有統(tǒng)計(jì)學(xué)意...
