【摘要】：為了研究不同劑量的重組人促紅細(xì)胞生成素(rhEpo)在Balb/c雌性小鼠上建立繼發(fā)性紅細(xì)胞增多癥小鼠模型的效果,為該病的新藥研發(fā)提供高效優(yōu)質(zhì)的服務(wù)平臺(tái),試驗(yàn)采用8~10周齡的Balb/c雌性小鼠,在試驗(yàn)前2~3天按體重隨機(jī)分為Saline-陰性對(duì)照組、rhEpo-3IU-4d組、rhEpo-10IU-4d組、rhEpo-10IU-7d組、rhEpo-10IU-14d組、rhEpo-30IU-4d組、rhEpo-10IU-4d 恢復(fù)期組,在最后一次皮下注射rhEpo 24 h后對(duì)試驗(yàn)小鼠進(jìn)行安樂死并采集血液和組織,檢測(cè)網(wǎng)織紅細(xì)胞數(shù)量、血細(xì)胞比容以及脾臟重量。結(jié)果表明:隨著rhEpo劑量和注射次數(shù)的增加,網(wǎng)織紅細(xì)胞數(shù)量、血細(xì)胞比容和脾臟重量都有顯著增加,但是對(duì)于動(dòng)物的體重并沒有太大影響。rhEpo-10IU-4d組網(wǎng)織紅細(xì)胞數(shù)量增加了5.9倍,血細(xì)胞比容增加了10%以上;rhEpo-10IU-14d組血細(xì)胞比容進(jìn)一步增加,但是網(wǎng)織紅細(xì)胞數(shù)量和脾臟重量未繼續(xù)增加。說明連續(xù)皮下注射不同劑量的rhEpo能夠誘導(dǎo)不同程度的繼發(fā)性細(xì)胞增多癥。
[Abstract]:In order to study the effect of different doses of recombinant human erythropoietin (rhEpo) on the establishment of secondary erythrocythemia mouse model in Balb/c female mice, and to provide a high-efficient and high-quality service platform for the development of new drugs for this disease, Balb/c female mice aged 8-10 weeks were randomly divided into Saline- negative control group, rhEpo-3IU-4d group, rhEpo-10IU-4d group, rhEpo-10IU-7d group, rhEpo-10IU-14d group and rhEpo-30IU-4d group on the 3rd day before the experiment. In the rhEpo-10IU-4d recovery group, 24 hours after the last subcutaneous injection of rhEpo, the mice were euthanized and the blood and tissues were collected. The reticulocyte count, hematocrit and spleen weight were measured. The results showed that the number of reticulocytes, hematocrit and spleen weight increased significantly with the increase of rhEpo dosage and injection times. In rhEPO-10IU-4d group, reticulocyte number increased 5.9 times and hematocrit increased by more than 10%. The hematocrit increased further in rhEpo-10IU-14d group, but the reticulocyte number and spleen weight did not continue to increase. It is suggested that continuous subcutaneous injection of different doses of rhEpo can induce different degrees of secondary hypercythemia.
