不同濃度氧暴露對早產(chǎn)兒外周血單個核細胞SIRT1通路的影響

【摘要】:目的觀察不同濃度及不同時間氧暴露對低體質量早產(chǎn)兒外周血單個核細胞(PBMCs)內沉默信息調節(jié)因子2相關酶1(SIRT1)通路相關蛋白表達的影響,探討氧暴露導致氧化應激損傷的機制。方法選取低體質量早產(chǎn)兒80例,入院時診斷為新生兒呼吸窘迫綜合征且需要立即氧療。根據(jù)吸氧濃度分為對照組、低濃度吸氧組、中濃度吸氧組、高濃度吸氧組各20例。對照組吸氧濃度Fi O=21%或以FiO_221%吸氧,但總吸氧時間24 h;低濃度吸氧組FiO_221%,吸氧時間24 h,或因病情需要FiO_230%的吸氧時間24 h;中濃度吸氧組FiO_2為30%~40%,吸氧時間24 h,或因病情需要FiO_240%的吸氧時間24 h;高濃度吸氧組為FiO_2≥40%且吸氧時間24 h。分別于生后0、7、14、28 d時,均經(jīng)橈動脈采血2 m L,分離PBMCs。采用激光共聚焦顯微鏡技術檢測PBMCs中活性氧簇(ROS)水平并進行SIRT1定位檢測。采用Western blot法檢測PBMCs內胞核SIRT1蛋白及PBMCs內SUMO特異性蛋白酶1(SENP1)、乙;痯53蛋白表達。結果隨著吸氧濃度的增加,生后住院時間的延長,與對照組相比,PBMCs內的ROS含量逐漸增加(P0.05),SIRT1轉位率逐漸增加(P0.05),胞核SIRT1蛋白水平明顯降低,PBMCs內SENP1蛋白水平增加(P0.05),乙;痯53蛋白水平增加(P0.05)。結論高濃度的氧暴露及長時間的氧療時間可導致低體質量早產(chǎn)兒PBMCs中產(chǎn)生大量的ROS,其機制可能與PBMCs中SENP1蛋白表達增加、SIRT1核轉位、乙;痯53增高導致的氧化應激損傷有關。
[Abstract]:Objective to investigate the effects of different concentrations and time of oxygen exposure on the expression of silencing signaling factor 2 related enzyme 1 (SIRT1) pathway related protein in peripheral blood mononuclear cells (PBMCs) of premature infants with low body mass. To investigate the mechanism of oxidative stress injury induced by oxygen exposure. Methods 80 premature infants with low body mass were diagnosed as neonatal respiratory distress syndrome and needed immediate oxygen therapy. According to the oxygen concentration, 20 cases were divided into control group, low concentration oxygen group, middle concentration oxygen inhalation group and high concentration oxygen inhalation group.
