E64d對(duì)活體缺氧缺血性腦損傷新生大鼠松果體的短期影響

【摘要】:目的:了解缺氧缺血性腦損傷(Hypoxic-ischemic brain damage,HIBD)模型中松果體的損傷情況,了解凋亡、自噬及壞死相關(guān)蛋白的改變,初步探討E64d在HIBD后松果體損傷修復(fù)中的短期作用及可能機(jī)制。 方法:采用7日齡新生SD(Sprague Dawley)大鼠,根據(jù)改良Rice-Vannucci法建立HIBD模型。將新生大鼠隨機(jī)分為E64d干預(yù)組(E64d組)、不干預(yù)組(DMSO組)、假手術(shù)組(Sham組),并選取HIBD后2h、6h、12h、24h、48h五個(gè)時(shí)間點(diǎn),通過(guò)Western blot法測(cè)定松果體Cleaved caspase-3、Beclin-1、Spectrin αⅡ蛋白濃度的變化;采用HE染色及免疫組織化學(xué)法觀察松果體組織Cleaved caspase-3、IL-21染色情況,從細(xì)胞層面了解松果體組織損傷情況并探討可能的機(jī)制。 結(jié)果:1、Western blot法提示:與Sham組比,DMSO組松果體細(xì)胞凋亡相關(guān)蛋白Cleaved caspase-3在6h明顯升高,12h達(dá)到高峰,至少持續(xù)至24h,到48h開(kāi)始下降(P0.05);與DMSO組比,E64d組Cleaved caspase-3在6h、12h左右出現(xiàn)下降(P0.05),但24h后已無(wú)統(tǒng)計(jì)學(xué)差異(P0.05)。免疫組化法提示:Cleaved caspase-3在間質(zhì)細(xì)胞及松果體細(xì)胞中均有表達(dá),與Sham組比,DMSO組Cleaved caspase-3在2h即表達(dá)明顯增強(qiáng),48h時(shí)仍高(P0.05);與DMSO組比,E64d組2h表達(dá)Cleavedcaspase-3明顯下降(P0.05),但48h已無(wú)作用(P0.05)。 2、與Sham組比,DMSO組自噬相關(guān)蛋白Beclin-1升高,在6h的時(shí)候出現(xiàn)統(tǒng)計(jì)學(xué)差異,24h達(dá)到高峰,至少持續(xù)至48h仍存在明顯升高(P0.05);與DMSO組比,E64d組松果體組織表達(dá)Beclin-1在24h出現(xiàn)顯著下降(P0.05),其余時(shí)間點(diǎn)無(wú)統(tǒng)計(jì)學(xué)差異(P0.05)。 3、與Sham組比,DMSO組松果體細(xì)胞質(zhì)膜骨架蛋白Spectrin αⅡ分解增多,除6h點(diǎn)外,各時(shí)間點(diǎn)上均有明顯升高,高峰在12h左右(P0.05);與DMSO組比,,使用E64d在一定時(shí)間內(nèi)減少了細(xì)胞骨架的分解,保護(hù)了質(zhì)膜完整性,在12h、24h存在統(tǒng)計(jì)學(xué)意義(P0.05),但之后壞死仍在上升,48h已無(wú)明顯差異(P0.05)。 4、免疫組化顯示松果體組織細(xì)胞間質(zhì)中存在IL-21,2h三組之間無(wú)差別(P0.05),與Sham組比,DMSO組表達(dá)的IL-21在48h的時(shí)候明顯升高(P0.05);與DMSO組比,E64d組松果體IL-21顯著下降(P0.05)。 結(jié)論:1、HIBD后松果體短期內(nèi)出現(xiàn)細(xì)胞損傷,表現(xiàn)為核溶解及碎裂,部分細(xì)胞分界模糊,并且牽涉到凋亡、自噬及壞死三種形式。 2、在本實(shí)驗(yàn)中,腹腔注射E64d對(duì)HIBD后活體松果體有著直接的影響。對(duì)松果體凋亡相關(guān)蛋白Cleaved caspase-3、細(xì)胞骨架蛋白Spectrin αⅡ影響較大,而對(duì)自噬相關(guān)蛋白Beclin-1的影響相對(duì)較小。該模型中,E64d早期能抑制凋亡,保護(hù)細(xì)胞質(zhì)膜完整性,但保護(hù)作用時(shí)間在24小時(shí)內(nèi)。E64d未能完全逆轉(zhuǎn)松果體組織的損傷進(jìn)程。 3、HIBD后松果體損傷可能存在機(jī)體免疫機(jī)制參與,機(jī)體可能藉由局部組織IL-21的上調(diào)參與損傷進(jìn)程,而E64d能明顯降低IL-21的表達(dá)。
