不同日齡新生大鼠缺氧缺血性腦損傷后腦組織谷氨酸含量變化的研究

【摘要】:目的:研究代表不同胎齡新生兒缺氧缺血性腦病(hypoxic ischemic encephalopathy, HIE)的模型鼠腦組織中谷氨酸(glutamate, Glu)含量變化的時(shí)間依從性及干預(yù)措施。方法:將128只新生SD大鼠按出生日齡(postnatal age, P)分為P2、P6、P12、P184組,每組32只;各組按不同干預(yù)措施分為缺氧缺血性腦損傷(hypoxic ischemic brain damage, HIBD)模型對(duì)照組(H)、α-氨基羥甲基惡唑丙酸(AMPA)干預(yù)組(A)、GYKI52466干預(yù)組(G)與空白對(duì)照組(N),每組8只。除N組外,各組參照Rice方法建立HIBD標(biāo)準(zhǔn)模型,A、G組于造摸后立即給藥。用藥6小時(shí)后采集標(biāo)本,檢測(cè)各組腦組織Glu含量,同時(shí)做病理學(xué)檢查。結(jié)果:各日齡組腦組織中Glu含量比較,Glu含量與日齡呈負(fù)相關(guān)(r=-0.81,p0.01),P2組含量最高,與P6、P12、P18組比較均有顯著性差異[(670.2±139.4)μmol/gprot.vs.(576±139) μmol/gprot.,(670.2±139.4)μmol/gprot.vs.(441.9±114.9) μmol/gprot.,(670.2±139.4)μmol/gprot.vs.(287.0±82.8) μmol/gprot.,Q=3.88,9.41,15.80,P0.05或P0.01]。各日齡組內(nèi)比較,G組腦組織中Glu含量明顯低于A組、H組和N組[(348.6±128.5)μmol/gprot. vs.(608.6±176.7)μmol/gprot.,(348.6±128.5)μmol/gprot.vs.(554.2±206.2)μmol/gprot.,(348.6±128.5)μmol/gprot.vs.(463.8±167.1) μmol/gprot.,Q=10.72,8.49,4.75,P均0.01]。病理學(xué)改變與G1u含量變化呈一致性。結(jié)論:代表不同胎齡新生兒腦發(fā)育水平的不同日齡新生大鼠腦組織中,Glu的含量具有時(shí)間依從性,與日齡呈負(fù)相關(guān);HIBD可導(dǎo)致Glu含量明顯增加而加重腦損傷;而GYKI52466可顯著降低Glu含量,從而發(fā)揮神經(jīng)保護(hù)作用,且胎齡越小,效果越明顯。故應(yīng)用GYKI52466早期干預(yù)可望有效防治因早產(chǎn)、窒息導(dǎo)致新生兒腦損傷。

Methods: 128 neonatal SD rats were divided into two groups according to their birth age (postnatal age, P): P2P6 P12P184 group with 32 rats in each group.
