ICAM-1、MCP-1表達(dá)水平及基因多態(tài)性與EV71手足口病

【摘要】:目的研究ICAM-1基因K469E位點(diǎn)、MCP-1A2518G位點(diǎn)基因多態(tài)性及sICAM-1、MCP-1在血清中表達(dá)水平與EV71手足口病的關(guān)系,探討EV71型手足口病的遺傳易感因素。方法收集手足口病患兒急性期(發(fā)病24小時(shí)內(nèi))大便標(biāo)本,常規(guī)處理大便標(biāo)本,運(yùn)用RT-PCR方法,采用腸道病毒71型(EV71)特異引物擴(kuò)增檢測(cè)病毒RNA。采用雙夾心抗體法(ELISA)檢測(cè)EV71感染陽(yáng)性組和對(duì)照組血清sICAM-1和MCP-1水平,并進(jìn)行直線相關(guān)分析。運(yùn)用限制性片段長(zhǎng)度多態(tài)性—聚合酶鏈反應(yīng)(PCR-RFLP)檢測(cè)EV71感染陽(yáng)性組和對(duì)照組中ICAM-1K469E位點(diǎn)及MCP-1A2518G位點(diǎn)堿基變異情況,并取樣采用堿基直接測(cè)序進(jìn)行驗(yàn)證,對(duì)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果①306例手足口病患兒大便標(biāo)本檢測(cè)中153例EV71陽(yáng)性,其中男83例,女70例,年齡11月~6歲,平均年齡2.5歲。②手足口病組患兒血清中sICAM-1和MCP-1水平均顯著高于正常對(duì)照組(P均0.01)。③EV71手足口病組和對(duì)照組ICAM-1K469E位點(diǎn)基因型分布存在顯著差異(x2=12.433,P0.01),EV71手足口病組A等位基因頻率明顯低于對(duì)照組(x2=8.073,P0.01)。④EV71手足口病組患兒MCP-1基因型分布、等位基因頻率與對(duì)照組比較均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論①ICAM-1和MCP-1表達(dá)水平與EV71手足口病有關(guān),推測(cè)sICAM-1參與機(jī)體EV71感染后的炎癥反應(yīng)和免疫損傷過(guò)程。②ICAM-1基因K469E位點(diǎn)多態(tài)性與EV71手足口病相關(guān),推測(cè)A等位基因是EV71感染的保護(hù)性基因,可降低EV71手足口病發(fā)生率。③MCP-1基因A-2518G位點(diǎn)基因多態(tài)性與EV71手足口病無(wú)關(guān)。
[Abstract]:Objective to study the relationship between the polymorphism of ICAM-1 gene K469E locus MCP-1A2518G and the expression level of sICAM-1 MCP-1 in serum with EV71 hand-foot-mouth disease (HFMD), and to explore the genetic susceptibility factors of EV71 Hand-foot-mouth disease (HMD). Methods stool specimens of children with hand-foot-mouth disease (HFMD) in acute phase (within 24 hours of onset) were collected. Stool specimens were treated with routine method. RT-PCR method was used to detect the viral RNA by using specific primers of enterovirus 71 (EV71). The serum levels of sICAM-1 and MCP-1 were detected by double sandwich antibody (ELISA) in the positive group of EV71 infection and the control group, and the linear correlation analysis was carried out. Restriction fragment length polymorphism (PCR-RFLP) was used to detect the variation of ICAM-1K469E and MCP-1A2518G loci in EV71 positive group and control group. The data were analyzed by direct sequencing. Results among the 1306 stool specimens of hand, foot and mouth disease, 83 were male and 70 were female. The age was 6 years old in November. The mean age of 2.5 years old in the HFMD group was significantly higher than that in the normal control group (P0.01). 3There was a significant difference in the distribution of ICAM-1K469E genotypes between the HFMD group and the control group (x2v12.433P0.01). The frequency of MCP-1 gene was significantly lower than that of control group (x2 + 8.073U, P0.01) .4EV71 Hand-foot-mouth disease (HFMD) group, the distribution of MCP-1 genotype was significantly lower than that in control group. There was no significant difference in allele frequency between the control group and the control group (P0.05). Conclusion the expression of 1ICAM-1 and MCP-1 is related to EV71 HFMD. It is speculated that sICAM-1 is involved in the inflammatory reaction and the process of immune damage after EV71 infection. The K469E polymorphism of K469E gene of ICAM-1 gene is associated with EV71 HFMD. It is suggested that A allele is a protective gene for EV71 infection, which can reduce the incidence of EV71 HFMD. 3The polymorphism of A-2518G locus of MCP-1 gene is not related to EV71 HFMD.
