咳喘寧口服液對哮喘模型大鼠中核因子-κB p65和TLR4的影響

【摘要】:目的:通過建立SD(Sprague-Dawley)大鼠哮喘模型,采用免疫組化的方法,檢測哮喘大鼠肺組織中核因子-κB p65及TLR4的表達(dá)水平,探討核因子-κB p65及TLR4在哮喘發(fā)病中的表達(dá)變化以及咳喘寧對其的影響。 方法:將50只SPF級雄性SD大鼠隨機(jī)分為:正常組,不做任何處理;模型組,用卵清蛋白誘發(fā)哮喘;咳喘寧組,使用卵清蛋白誘發(fā)哮喘后,用中藥咳喘寧口服液治療;布地奈德組,用卵清蛋白誘喘后用布地奈德治療。采用免疫組化法檢測各組肺組織中核因子-κB p65及TLR4的水平;采用全自動血細(xì)胞計(jì)數(shù)儀對肺泡灌洗液中白細(xì)胞總數(shù)、中性粒細(xì)胞、嗜酸性粒細(xì)胞(EOS)及淋巴細(xì)胞計(jì)數(shù);并觀察支氣管肺組織的病理形態(tài)學(xué)改變。 結(jié)果: 1.各組大鼠支氣管肺泡洗灌液的細(xì)胞計(jì)數(shù)及分類計(jì)數(shù):模型組BALF中白細(xì)胞總數(shù)、中性粒細(xì)胞數(shù)、淋巴細(xì)胞數(shù)及EOS均升高(P0.05);咳喘寧組與模型組相較,BALF中白細(xì)胞總數(shù)、淋巴細(xì)胞數(shù)、EOS均降低(P0.05);咳喘寧組與布地奈德組相較,BALF中各細(xì)胞數(shù)無明顯差異(P0.05)。 2.各組大鼠肺組織病理變化:正常組無明顯炎癥改變。模型組呈顯著炎性變化。布地奈德組與咳喘寧組均呈輕度炎癥變化。 3.各組大鼠肺組織核因子-κB p65免疫組化表達(dá):核因子-κB p65在正常組大鼠肺組織呈弱陽性表達(dá);模型組大鼠肺組織內(nèi)核因子-κBp65的表達(dá)明顯較正常組增加;布地奈德組和咳喘寧組核因子-κB p65的表達(dá)強(qiáng)于正常組,弱于模型組,布地奈德組與咳喘寧組比較核因子-κB p65表達(dá)無明顯差異。 4.各組大鼠肺組織TLR4免疫組化表達(dá):TLR4在正常組大鼠肺組織呈弱陽性表達(dá);模型組大鼠肺組織內(nèi)TLR4的表達(dá)明顯較正常組增加;布地奈德組和咳喘寧組TLR4的表達(dá)強(qiáng)于正常組,弱于模型組,布地奈德組與咳喘寧組比較TLR4表達(dá)無明顯差異。 結(jié)論:咳喘寧口服液能減輕哮喘大鼠肺組織的炎癥反應(yīng);模型組大鼠肺組織中TLR4、NF-κB P65的陽性表達(dá)明顯升高,咳喘寧口服液治療哮喘能降低TLR4、NF-κB P65的表達(dá)。
[Abstract]:Objective: to study the expression of nuclear factor- 魏 B p65 and TLR4 in lung tissue of SD rague-Dawley (SD) rats by immunohistochemical method. To investigate the expression of nuclear factor- 魏 B p65 and TLR4 in asthma and the effect of Kechuanning on them. Methods: fifty SPF male SD rats were randomly divided into normal group without any treatment, model group with ovalbumin induced asthma and Kechuanning group with Kechuanning oral liquid. Budesonide group was treated with budesonide after ovalbumin induced asthma. The levels of nuclear factor- 魏 B p65 and TLR4 in lung tissue were detected by immunohistochemical method, and the total number of leukocytes, neutrophils, eosinophils and lymphocytes in alveolar lavage fluid were counted by automatic blood cell counter. The histopathological changes of bronchopulmonary tissue were observed. Results: 1. The cell count and classification count of bronchoalveolar lavage fluid in each group: the total number of leukocytes, neutrophils, lymphocytes and EOS in BALF of model group were all increased (P 0.05), and the total number of leukocytes in Kechuanning group was higher than that in model group. In Kechuanning group and budesonide group, there was no significant difference in the number of cells in BALF between Kechuanning group and budesonide group. 2. Pathological changes of lung tissue in each group: there was no obvious inflammatory change in normal group. The model group showed significant inflammatory changes. Both budesonide group and Kechuanning group showed mild inflammation. 3. The expression of nuclear factor- 魏 B p65 was weakly positive in the normal group, and the expression of nuclear factor- 魏 B p65 in the model group was significantly higher than that in the normal group. The expression of NF- 魏 B p65 in budesonide group and Kechuanning group was higher than that in normal group, but weaker than that in model group. There was no significant difference in the expression of NF- 魏 B p65 between budesonide group and Kechuanning group. 4. The expression of TLR4 was weakly positive in the normal group, the expression of TLR4 in the model group was significantly higher than that in the normal group, and the expression of TLR4 in the budesonide group and Kechuanning group was stronger than that in the normal group. There was no significant difference in TLR4 expression between budesonide group and Kechuanning group. Conclusion: Kechuanning oral liquid can attenuate the inflammatory reaction of lung tissue in asthmatic rats, and the positive expression of TLR4 and NF- 魏 B p65 in lung tissue of model group is obviously increased, while Kechuanning oral liquid can reduce the expression of TLR4- 魏 B p65 in asthmatic rats.
