江浙滬地區(qū)EV71的分子流行病學(xué)特征

【摘要】:目的研究江浙滬地區(qū)EV71-VP1遺傳變異特征,了解江浙滬地區(qū)EV71病毒基因突變及抗原變異規(guī)律,為EV71的預(yù)防、治療提供幫助。方法從GenBank上下載了江浙滬地區(qū)2009-2014年共79株EV71的VP1區(qū)段序列與各亞型參考株序列,用MEGA7.0軟件構(gòu)建進(jìn)化樹,分析江浙滬地區(qū)EV71-VP1分子流行病學(xué)特征;用疫苗株FY23株做為參考株比較江浙滬地區(qū)EV71毒株的氨基酸變異情況。結(jié)果通過進(jìn)化樹與序列同源分析,發(fā)現(xiàn)79株毒株均為C4a亞型,江浙滬地區(qū)EV71毒株可分為兩個(gè)大簇與數(shù)個(gè)小簇,兩個(gè)大簇間沒有發(fā)現(xiàn)明顯的序列差異,江浙滬三個(gè)地區(qū)的毒株在進(jìn)化樹上的分布不存在明顯的差異。79株毒株共存在34個(gè)氨基酸位點(diǎn)突變,其中與病毒致病及疫苗接種效果相關(guān)的98、145、289位點(diǎn)發(fā)生多株突變,7株發(fā)生了病毒與受體結(jié)合相關(guān)的E145G/Q突變,7株發(fā)生了位于病毒表面BC環(huán)內(nèi)的E98K突變,5株發(fā)生了與臨床癥狀表現(xiàn)相關(guān)的A289T/V突變,未發(fā)現(xiàn)2009-2014年有明顯的EV71毒株氨基酸變異差異。結(jié)論雖然2009-2014年江浙滬地區(qū)的EV71毒株流行亞型沒有改變,也不存在明顯地區(qū)間的流行差異,但是病毒的基因及氨基酸突變較為多見,仍應(yīng)關(guān)注該地區(qū)的病毒基因及抗原變異,以便更好的預(yù)防EV71疫情。
[Abstract]:Objective to study the genetic variation characteristics of EV71-VP1 in Jiangsu, Zhejiang and Shanghai, and to understand the rules of gene mutation and antigen variation of EV71 virus in Jiangsu, Zhejiang and Shanghai, so as to prevent EV71. Methods from 2009 to 2014, 79 EV71 strains from Jiangsu, Zhejiang and Shanghai were downloaded from the GenBank to construct the evolutionary tree of each subtype of EV71. The molecular epidemiological characteristics of EV71-VP1 in Jiangsu, Zhejiang and Shanghai were analyzed. The amino acid variation of EV71 strains in Jiangsu, Zhejiang and Shanghai regions was compared by using vaccine FY23 strain as reference strain. Results 79 strains were found to be C4a subtype by phylogenetic tree and sequence homology analysis. The EV71 strains in Jiangsu, Zhejiang and Shanghai can be divided into two large clusters and several small clusters, and there is no obvious sequence difference between the two clusters. There was no significant difference in the distribution of virus strains in evolutionary trees between Jiangsu, Zhejiang and Shanghai. There were 34 amino acid loci mutations in 79 strains. Among them, there were several mutations at the 98145289 site associated with virus pathogenicity and inoculation effect. 7 strains of E145G/Q mutation associated with virus receptor binding occurred and 5 strains of E98K mutation located in the BC loop on the surface of the virus were found to be associated with clinical practice. A289T/V mutations associated with symptoms, No significant amino acid variation of EV71 strains was found in 2009-2014. Conclusion although the epidemic subtypes of EV71 strains in Jiangsu, Zhejiang and Shanghai regions have not changed from 2009 to 2014, there are no significant differences between regions, but the gene and amino acid mutations of the virus are more frequent. Attention should still be paid to the genetic and antigenic variation of the virus in order to better prevent the outbreak of EV71.
