【摘要】：目的研究磷酸二酯酶4D(PDE4D)基因rs1544791位點多態(tài)性與中國漢族兒童支氣管哮喘的相關(guān)性。方法采用病例-對照的研究方法,選取2014年1月至2015年12月在青島大學附屬醫(yī)院就診的132名中國漢族支氣管哮喘患兒及181名健康查體兒童作為研究對象,抽取外周靜脈血,應(yīng)用DNA提取試劑盒提取外周血白細胞DNA,應(yīng)用聚合酶鏈反應(yīng)(PCR)擴增,經(jīng)外切核酸酶及蝦堿酶純化后,用連接酶反應(yīng)連接熒光標記的特異探針,通過毛細管電泳來區(qū)分基因位點,并對比分析出兩組人群的基因型頻率和等位基因頻率的差異,應(yīng)用卡方檢驗對結(jié)果進行比較分析。結(jié)果哮喘組和對照組PDE4D基因rs1544791位點的分布均符合Hardy-Weinberg平衡定律,兩組PDE4Drs1544791位點的3種基因型(TT、CT、CC)的分布及等位基因頻率與正常對照組相比差異均無統(tǒng)計學意義(p0.05)。結(jié)論磷酸二酯酶4D(PDE4D)基因rs1544791位點多態(tài)性與中國漢族兒童支氣管哮喘無相關(guān)性。
[Abstract]:Objective to study the association between the rs1544791 polymorphism of phosphodiesterase 4D (PDE4D) gene and bronchial asthma in Chinese Han children. From January 2014 to December 2015, 132 children with bronchial asthma of Han nationality and 181 healthy children were selected as study subjects, and peripheral venous blood was drawn. The peripheral blood leukocyte DNA was extracted by DNA extraction kit and amplified by polymerase chain reaction (PCR). After purified by exonuclease and shrimp base enzyme, ligase reaction was used to connect the specific probe labeled with fluorescence, and the loci were identified by capillary electrophoresis. The differences of genotype frequency and allele frequency between the two groups were compared and analyzed by chi-square test. Results the distribution of rs1544791 loci of PDE4D gene in asthmatic group and control group was in accordance with the law of Hardy-Weinberg equilibrium. There was no significant difference in the distribution and allelic frequency of the three genotypes of PDE4Drs1544791 between the two groups compared with the control group (P 0.05). Conclusion there is no correlation between the polymorphism of rs1544791 locus of phosphodiesterase 4DG PDE4D gene and bronchial asthma in Chinese Han children.
