巨細(xì)胞病毒感染后新生小鼠腦內(nèi)Wif-1、Shh的表達(dá)變化

【摘要】:目的:探討鼠巨細(xì)胞病毒(murine cytomegalovirus,MCMV)感染后新生小鼠腦內(nèi)Wnt inhibitory factor-1(Wif-1)、Sonic hedgehog(Shh)信號(hào)分子的表達(dá)變化及對(duì)中樞神經(jīng)系統(tǒng)發(fā)育的可能影響。方法:體外3T3細(xì)胞培養(yǎng)、MCMV接種增殖;60只生后3 d的BALB/c仔鼠隨機(jī)分為正常對(duì)照組及病毒感染組,病毒組通過(guò)腹腔注射法建立新生小鼠中樞神經(jīng)系統(tǒng)MCMV感染模型,而對(duì)照組則注射等量的生理鹽水;每組設(shè)立3個(gè)時(shí)間點(diǎn)亞組(造模后3 d,7 d,14 d),相應(yīng)時(shí)點(diǎn)處死仔鼠,取腦組織備用。通過(guò)RT-PCR檢測(cè)兩組各時(shí)點(diǎn)腦組織中的MCMV-DNA表達(dá);Western Blot檢測(cè)小鼠腦組織中Wif-1、Shh蛋白的表達(dá);流式細(xì)胞術(shù)檢測(cè)兩組的細(xì)胞周期比例改變。結(jié)果:病毒組小鼠腦組織中MCMV-DNA的表達(dá)隨著時(shí)間推移持續(xù)增多,而對(duì)照組3個(gè)時(shí)間點(diǎn)均陰性表達(dá);病毒組小鼠腦組織中Wif-1分子的表達(dá)相對(duì)于對(duì)照組增強(qiáng),而Shh分子的表達(dá)相比于對(duì)照組則表現(xiàn)出明顯的抑制;病毒組小鼠腦組織中處于分裂期的細(xì)胞百分比較對(duì)照組下降。結(jié)論:MCMV感染后的新生小鼠腦組織內(nèi)Wif-1表達(dá)增強(qiáng),而Shh信號(hào)分子則下調(diào),其表達(dá)異常變化可能影響Wnt、Hedgehog信號(hào)通路,并影響中樞神經(jīng)系統(tǒng)成熟過(guò)程。
[Abstract]:Objective: to study murine cytomegalovirus of murine cytomegalovirus. Wnt inhibitory factor-1 and Wif-1 were found in the brain of newborn mice after MCMV infection. Changes of signal molecule expression in Sonic hedgehog and its possible effect on the development of central nervous system methods: 3T3 cells cultured in vitro were inoculated and proliferated. Sixty postnatal BALB/c mice were randomly divided into normal control group and viral infection group. The MCMV infection model of central nervous system of newborn mice was established by intraperitoneal injection in the virus group. The control group was injected with the same amount of normal saline. Each group was divided into 3 subgroups (3 days, 7 days and 14 days after modeling), and the rats were killed at the corresponding time points. RT-PCR was used to detect the expression of MCMV-DNA in brain tissue at different time points. Western Blot was used to detect the expression of Wif-1 protein in mouse brain. Results: the expression of MCMV-DNA in the brain tissue of mice in the virus group increased with the passage of time, but the expression of MCMV-DNA in the control group was negative at three time points. Compared with the control group, the expression of Wif-1 in the brain tissue of the virus group was increased, while the expression of the Shh molecule was significantly inhibited compared with the control group. Compared with the control group, the percentage of cells in the brain tissue of the virus group decreased. Conclusion the expression of Wif-1 in the brain tissue of newborn mice infected with MCMV is increased, while the Shh signal molecule is down-regulated. The abnormal changes of its expression may affect the Hedgehog signaling pathway and the maturation process of the central nervous system (CNS).
