squamous cellcarcinoma,)組織中的表達(dá)及與臨床病理的關(guān)系;觀察藍(lán)萼甲素(Glaucocalyxin A)對宮頸癌SiHa細(xì)胞的作用并探討其機(jī)制。方法:組織標(biāo)本89例及正常宮頸上皮(normal cervical epithelium,NCE)組織標(biāo)本25例,采用免疫組織化學(xué)法,組織及NCE組織中PDCD5、Bcl.2蛋白的表達(dá)情況;四***偶氮唑鹽(methyl thiazolyl tetrazolium,MTT)比色法觀察1.25pmol/L、2.51amol/L、5pmol/L、lOmnol/L、20pmol/L的Glaucocalyxin A對SiHa細(xì)胞增殖的影響;Hoechst33258熒光染色和透射電鏡(transmission electron microscopy,TEM)觀察給藥后SiHa細(xì)胞的形態(tài)學(xué)變化:流式細(xì)胞儀(flow cytometer,FCM)檢測給藥后SiHa細(xì)胞周期和凋亡率的變化;RT-PCR檢測給藥后SiHa細(xì)胞凋亡相關(guān)基因PDCD5、Caspase.3、Caspase.9、p53、Bcl-2、Bax mRNA表達(dá)的變化;Western blot檢測給藥后SiHa細(xì)胞凋亡相關(guān)蛋白PDCD5、Bcl.2、Bax蛋白的表達(dá)情況。結(jié)果:組織中PDCD5蛋白的陽性表達(dá)率為33.7l%(30/89)低于NCE組織的76%(19/25),PDCD5蛋白的陽性表達(dá)率與有無淋巴結(jié)轉(zhuǎn)移極顯著相關(guān)(P<0.001),與病理分級低分化和高分化顯著相關(guān)(尸<0.05)。組織中Bcl.2蛋白的陽性表達(dá)率為67.42%(60/89)高于NCE組織的40%(10/25),Bcl.2蛋白的陽性表達(dá)率與宮頸癌的浸潤深度極顯著相關(guān)(氏0.001),與腫瘤的病理分級及有無淋巴結(jié)轉(zhuǎn)移非常顯著相關(guān)(P<O.01)。組織中Bcl.2和PDCD5蛋白表達(dá)呈負(fù)相關(guān)(產(chǎn)一0.427,P<O.01);藍(lán)萼甲素對SiHa細(xì)胞有抑制增殖效應(yīng),且呈劑量及時(shí)間依賴趨勢;Hoechst 33258染色后,熒光顯微鏡下見SiHa細(xì)胞的胞核致密濃染,或里碎塊狀;TEM觀察顯示SiHa細(xì)胞出現(xiàn)細(xì)胞核皺縮,染色質(zhì)邊集等細(xì)胞凋亡的特征性改變;FCM檢測結(jié)果,隨著時(shí)間的延長,SiHa細(xì)胞凋亡率逐漸增加且S期細(xì)胞比例也逐漸增加;RT-PCR結(jié)果顯示Bcl.2 mRNA的表達(dá)下調(diào),PDCD5、Caspase.3、Caspase.9、p5 3、BaxmRNA的表達(dá)上調(diào);Western molecular mechanisms involved in theeffects ofGlaucocalyxin Aon SiHa cells were studied by RT-PCR and Western blot.Results There were significant differences of PDCD5 expression between tissues(33.71%,30/89)and NCE tissues(76%,19/25).The expression rate Wasassociated wim pathological grade(P<0.05)and lymph node metastasis(P<0.00 1).There were also significant differences of Bcl-2
