高含油樟樹無性系內生菌多樣性及篩選

目前對樟樹(Cinnamomum camphora(L.)Presl.)的研究主要集中在精油成分分析、扦插育苗技術和矮化造林等方面,尚未深入微生物層面分析,尤其探索樟樹內生菌分離、鑒定及與宿主植物及次生產物生長發(fā)育之間的聯系更是尚未涉獵。因此,本研究采用16srDNA高通量測序技術對三個季節(jié)的樣品的內生菌進行測序,研究樟樹在不同季節(jié)(生長期)的生理生化、內生菌群落多樣性及次生代謝,同時篩選出促生樹樟精油的優(yōu)勢內生菌,為研究樟樹內生菌的功能及其應用提供理論基礎。本研究通過對5個樟樹無性系葉片的生理特性、生化指標和精油含量及精油成分來探究內生菌與樟樹的生理生化及精油(含量和成分)的相關性。并且采用組織塊分離法,主要通過顯微鏡觀察,分離純化樟樹內生細菌和真菌。使用分子鑒定策略進一步分析不同無性系內生菌多樣性、代表性細菌和真菌。通過固相微萃取純化內生菌之后,提取和分離的代謝產物,測定代謝產物成分,并與樟樹葉精油成分進行比對,來篩選促進樟樹葉精油含量的內生菌。獲得的結果如下。在不同的采樣時間,獲得了來自葉子的10,844,124個有效序列。Shannon指數表明,在采樣期間,細菌群落的多樣性較低...
