桔小實(shí)蠅發(fā)育階段轉(zhuǎn)變前后miRNA分析及miR-309a調(diào)控卵巢發(fā)育的分子機(jī)制

microRNA(miRNA)是一類廣泛存在于生物體內(nèi)長(zhǎng)度約22 bp的內(nèi)源性小分子非編碼RNA,在轉(zhuǎn)錄后水平調(diào)控基因的表達(dá)。miRNA因參與調(diào)控昆蟲的生長(zhǎng)發(fā)育、生殖、免疫和抗藥性等諸多生理過程,近年來逐漸成為昆蟲發(fā)育生物學(xué)領(lǐng)域的一個(gè)研究熱點(diǎn)。桔小實(shí)蠅Bactrocera dorsalis是一種危險(xiǎn)性的果蔬害蟲,寄主范圍廣,造成的經(jīng)濟(jì)損失嚴(yán)重,多個(gè)國(guó)家和地區(qū)將其列為檢疫性對(duì)象。目前,桔小實(shí)蠅已成為我國(guó)果蔬產(chǎn)業(yè)健康發(fā)展的巨大威脅,探索桔小實(shí)蠅新的防控策略顯得尤為迫切。據(jù)此,本學(xué)位論文以桔小實(shí)蠅為研究對(duì)象,基于不同發(fā)育階段轉(zhuǎn)變前后高通量測(cè)序,在注釋和鑒定miRNA的基礎(chǔ)上篩選差異表達(dá)的miRNA;利用qRT-PCR,解析卵和蛹期與發(fā)育相關(guān)的miRNA表達(dá)譜,同時(shí)結(jié)合使用miRNA類似物/抑制劑探究miRNA參與調(diào)控化蛹及羽化進(jìn)程的功能;最后聚焦miR-309a,綜合運(yùn)用qRT-PCR、靶基因預(yù)測(cè)、雙熒光素酶報(bào)告系統(tǒng)、RNA結(jié)合蛋白免疫沉淀和外源激素處理等技術(shù)和方法,初步提出JH-miR-309a-pnr-Vg related genes通路調(diào)控桔小實(shí)蠅生殖的分子機(jī)理。主要研究結(jié)果如下:1桔...
